Curcumin possesses strong antioxidant capacities, which may explain its effects against degenerative diseases in which oxidative stress plays a major role. As previously described for flavonoids, it is unlikely that curcumin acts as a direct antioxidant outside the digestive tract since its concentration in peripheral blood and organs is very low (near or below 1 pM, even after acute or long-term supplementation). The anticancer effect of curcumin in murine thymoma cells was found to be due to the blocking of IL-1 signaling by the inhibition of the recruitment of the IL-1 receptor-associated kinase (IRAK) [Jurrmann et al., 2005]. In another clinical trial, researchers evaluated the effect of oral curcumin with piperine on the pain, and the markers of oxidative stress in patients with tropical pancreatitis (TP). Twenty patients with tropical pancreatitis were randomized to receive 500 mg of curcumin with 5 mg of piperine, or placebo, for 6 weeks, and the effects on the pattern of pain, and on red blood cell levels of malonyldialdehyde (MDA) and glutathione (GSH) were assessed. There was a significant reduction in the erythrocyte MDA levels following curcumin therapy compared with placebo with a significant increase in GSH levels.
