Clobetasol 17-propionate

Chloro-9-fluoro-llb-hydroxy-16b-methyl-17-(l-oxopropoxy)-pregna-l,4-diene-3,20-dione. See Clobetasol propionate. [Pg.199]

Activity is also retained when the hydroxyl group at the 21 position is replaced by chlorine. Reaction of corticoid 44 with methanesulfonyl chloride proceeds preferentially at the 21-hydroxyl (45) due to the hindered nature of the 11-alcohol. Replacement of the mesylate by means of lithium chloride in DMF affords clobetasol propionate (46) a similar sequence starting with the 17- butyrate ester 47, via mesylate 48, should give clobetasone butyrate, (49) [11]. [Pg.72]

Examples of group I, i.e. weak or low efficacy topical steroids, are hydrocortisone acetate in various concentrations, methylprednisolone 1.0% and prednisolone 0.5%. Group II, the moderately potent steroids, includes alclometasone dipropionate 0.05%, hydrocortisone butyrate 0.1%, triamcinolone acetonide 0.025% and fluocinolone ace-tonide 0.01%. Group III, the potent steroids, contains among others betamethasone valerate 0.1%, betamethasone dipropionate 0.05%, budesonide 0.025%, desoximetasone 0.05%, fluticasone propionate 0.05%, amcinonide 0.1%, fluocinonide 0.05% and mometasone furoate 0.1%. Group IV comprises the very potent agents such as clobetasol propionate 0.05% and halobetasol propionate 0.05%. [Pg.483]

Topical preparations usually contain relatively insoluble steroids, such as clobetasol propionate, triamcinolone acetonide, or triamcinolone diacetate. Side effects of this mode of drug application are usually milder and more transient than those seen after systemically administered steroids. However, potent topical corticosteroids, such as clobetasol propionate (Temovate), can suppress adrenal function when used in large amounts for a long time, especially when the skin surface is denuded or when occlusive dressings are employed. Since the high potency topical preparations carry a higher risk of local side effects, their use should be held in reserve. [Pg.692]

Overdosage can occur from topically applied clobetasol propionate absorbed in sufficient amounts to produce systemic effects producing reversible adrenal suppression, manifestations of Cushi ng s syndrome, hyperglycemia, and glucosuria in some patients. [Pg.281]

Steroid creams will often clear the condition and hydrocortisone can be prescribed under a variety of trade names such as Alphaderm. More powerful are the creams based on betamethasone esters, of which Betnovate is the best known and has been used for more than 40 years. If this does not work then the doctor can prescribe even stronger drugs based on clobetasol propionate, such as Dermovate, or similar active agents. There are several such creams now available and it is rare for eczema not to respond to one of them and be cured - and kept at bay. If the eczema is infected then various antibiotics, antivirals, and anti-fungals can be given to clear up the infection. Very... [Pg.43]

The anti-inflammatory and antihypoproliferative activities of this compound were compared to those of other compounds in several animal models. The compound was 1300-fold more potent than hydrocortisone, and 11-fold more potent than clobetasol propionate, respectively, in suppressing the erythemal responses in a guinea pig model of UV-induced inflammation, and it was 10-fold more potent than clobetasol in the cotton pellet granuloma assay in rats. [Pg.432]
