Anti-fungals

A cyclocondensation involving the carbonyl of a benzoquinone was used for the synthesis of a cinnoline 15 of interest as a potential anti-fungal agent <06BMCL1850>. 

Stability constants for a series of M2+ cations, including Ca2+, with A23187 and with ionomycin have been determined in methanol-water mixtures (Taylor, R. W. Pfeiffer, D. R. Chapman, C. J. Craig, M. E. Thomas, T. P. Pure Appl. Chem. 1993, 65, 579-584) the estimation of stability constants for complexes of the anti-fungal antibiotic pradimicin (BMY-28864) with alkaline earth cations is complicated by the tendency of this antibiotic to aggregate (Hu, M. Ishizuka, Y. Igarashi, Y. Oki, T. Nakanishi, H. Spectrochim. Acta A 1999, 56, 181-191).

Moustafa and Ahmed reported the synthesis of isoxazoles [92,93]. These compounds are related to pyrazoles except that an oxygen replaces the amine nitrogen. Scheme 49 shows the synthesis of 182 from the corresponding chal-cone 183 by reaction with hydroxylamine. Although these compounds were not tested for cytotoxicity as tubulin binders, they were found to possess antibacterial and anti-fungal activity. 

In 2003, Sunthitikawinsakul et al. described the anti-mycobacterial activity of various 3-methylcarbazole derivatives, including 3-formylcarbazole (3), methyl carbazole-3-carboxylate (4) (see Scheme 2.2), clausine K (clauszoline-J) (51) (see Scheme 2.11), and 7-methoxymukonal (68) (see Scheme 2.14) against Mycobacterium tuberculosis H37Ra. Except for clausine K (clauszoline-J) (51), all of these alkaloids also showed anti-fungal activity against Candida albicans (442). In 2005, Eranzblau et al. reported the in vitro anti-TB activity of various carbazole derivatives such as 3-formylcarbazole (3), methyl carbazole-3-carboxylate (4) (see Scheme 2.2), lansine...

There are occasional anomalies to the rule that food reduces and delays peak plasma concentration. The anti-fungal drug, griseofulvin, has enhanced absorption if taken with a meal - possibly because it becomes emulsified by bile salts and passes more readily into the lymphatic drainage of the gut which bypasses the liver, entering the venous system directly. The immuno-suppressant cyclosporin, and calcium salts in general, show a similar increase in absorption when taken with a fatty meal.
