MDCK (Madin-Darby canine kidney) cells are derived from distal tubules, whereas LLC-PKi are from proximal tubes. b BMEC (brain microvessel endothelial cells) are isolated from capillaries. BPAEC (bovine pulmonary artery endothelial cells), BAEC (bovine aortic endothelial cells), and HUVEC (human umbilical vein endothelial cells) are large vessel endothelia. [Pg.241]

Calcium antagonists are able to affect nitric oxide production and suppress the peroxyni-trite-induced damage. Thus, nifedipine enhanced the bioavailability of endothelial NO in porcine endothelial cell cultures supposedly through an antioxidative mechanism [288], Pretreatment with nisoldipine, a vascular-selective calcium blocker of dihydropyridine-type, of confluent bovine aortic endothelial cells suppressed the peroxynitrite-induced GSH loss and increased cell survival [289]. [Pg.884]

Endothelial NOS was found to be associated with cell membranes and this property was associated with the finding that the cDNA contained the structure for binding myristic acid55. Busconi and Michel56 used bovine aortic endothelial cells to demonstrate that the myristoylated protein was found. When the second amino acid, glycine, was converted to alanine, the addition of myristic acid was prevented and the enzyme remained soluble. Sessa and coworkers51 carried out the same experiments simultaneously and found the same results. However, in both cases the enzyme was not purified, although the protein... [Pg.977]

Shown in Figure 6 is in vitro cytotoxic activity of PIPAAm-PBMA micelles loaded with ADR or micelles without ADR at 29°C (below the LCST) and at 37°C (above the LCST) compared with that of free ADR. In vitro cytotoxic activity was measured using bovine aorta endothelial cells. Bovine aortic endothelial cells were obtained as previously reported using dispase for cell dissociation from freshly harvested bovine aorta [13]. The cells plated at a density of 3 x cells/well, were exposed with free ADR or micelles loaded with ADR at below and above the LCST for 5 days. In order to assay cytotoxicity of the free ADR or micelles loaded with ADR, culture medium was replaced with 10% FBS-supplemented phenol red-free DMEM containing 10% alamar Blue, a dye that is subject to reduction by cytochrome c activity and changes the color from blue to red [38]. After 4-hour incubation, reduction of the dye was estimated by absorbance at 560 and 600 nm. PIPAAm-PBMA polymeric micelles loaded with ADR showed higher cytotoxic activity than that of free ADR at 37°C (above the LCST)... [Pg.43]

Okano et al. [51] applied poly(IPAAm) to modifying the surface of commercial polystyrene1 culture dishes for bovine aortic endothelial cells as well as for rat hepatocytes. Graft polymerization of IPAAm onto the polystyrene dishes was carried out by using an electron beam. The thickness of the poly(IPAAm)-grafted layer in aqueous systems was 0.5 pm at 37 °C and 0.6 pm at 15 °C. After endothelial cells were cultured at 37 °C for 2 days, the temperature was decreased to 10 °C and the number of cells that were detached from the surface was counted. Okano et al. observed that 100% of cells were detached from the poly(IPAAm)grafted surface. [Pg.20]
