Bilayer vesicles phase transitions

Phospholipid molecules form bilayer films or membranes about 5 nm in thickness as illustrated in Fig. XV-10. Vesicles or liposomes are closed bilayer shells in the 100-1000-nm size range formed on sonication of bilayer forming amphiphiles. Vesicles find use as controlled release and delivery vehicles in cosmetic lotions, agrochemicals, and, potentially, drugs. The advances in cryoelec-tron microscopy (see Section VIII-2A) in recent years have aided their characterization [70-72]. The other class of phenomenological approaches subsumes the random surface theories (Sec. B). These reduce the system to a set of internal surfaces, supposedly filled with amphiphiles, which can be described by an effective interface Hamiltonian. The internal surfaces represent either bilayers or monolayers—bilayers in binary amphiphile—water mixtures, and monolayers in ternary mixtures, where the monolayers are assumed to separate oil domains from water domains. Random surface theories have been formulated on lattices and in the continuum. In the latter case, they are an interesting application of the membrane theories which are studied in many areas of physics, from general statistical field theory to elementary particle physics [26]. Random surface theories for amphiphilic systems have been used to calculate shapes and distributions of vesicles, and phase transitions [27-31]. [Pg.639]

The development of monoalkyl phosphate as a low skin irritating anionic surfactant is accented in a review with 30 references on monoalkyl phosphate salts, including surface-active properties, cutaneous effects, and applications to paste and liquid-type skin cleansers, and also phosphorylation reactions from the viewpoint of industrial production [26]. Amine salts of acrylate ester polymers, which are physiologically acceptable and useful as surfactants, are prepared by transesterification of alkyl acrylate polymers with 4-morpholinethanol or the alkanolamines and fatty alcohols or alkoxylated alkylphenols, and neutralizing with carboxylic or phosphoric acid. The polymer salt was used as an emulsifying agent for oils and waxes [70]. Preparation of pharmaceutical liposomes with surfactants derived from phosphoric acid is described in [279]. Lipid bilayer vesicles comprise an anionic or zwitterionic surfactant which when dispersed in H20 at a temperature above the phase transition temperature is in a micellar phase and a second lipid which is a single-chain fatty acid, fatty acid ester, or fatty alcohol which is in an emulsion phase, and cholesterol or a derivative. [Pg.611]

The phase transition of bilayer lipids is related to the highly ordered arrangement of the lipids inside the vesicle. In the ordered gel state below a characteristic temperature, the lipid hydrocarbon chains are in an all-trans configuration. When the temperature is increased, an endothermic phase transition occurs, during which there is a trans-gauche rotational isomerization along the chains which results in a lateral expansion and decrease in thickness of the bilayer. This so-called gel to liquid-crystalline transition has been demonstrated in many different lipid systems and the relationship of the transition to molecular structure and environmental conditions has been studied extensively. [Pg.294]

Polymeric phospholipids based on dioctadecyldimethylammonium methacrylate were formed by photopolymerization to give polymer-encased vesicles which retained phase behavior. The polymerized vesicles were more stable than non-polymerized vesicles, and permeability experiments showed that vesicles polymerized above the phase transition temperature have lower permeability than the nonpolymerized ones [447-449]. Kono et al. [450,451] employed a polypeptide based on lysine, 2 aminoisobutyric acid and leucine as the sensitive polymer. In the latter reference the polypeptide adhered to the vesicular lipid bilayer membrane at high pH by assuming an amphiphilic helical conformation, while at low pH the structure was disturbed resulting in release of the encapsulated substances. [Pg.37]

A similar study by O Brien and coworkers utilized bilayers composed of a shorter chain diacetylenicPC (9) and DSPC or DOPC [37]. Phase separation was demonstrated in bilayers by calorimetry and photopolymerization behavior. DSC of the 9/DSPC (1 1) bilayers exhibited transitions at 40 °C and 55 °C, which were attributed to domains of the individual lipids. Polymerization at 20 °C proceeded at similar rates in the mixed bilayers and pure 9 bilayers. A dramatic hysteresis effect was observed for this system, if the bilayers were first incubated at T > 55 °C then cooled back to 20 °C, the DSC peak for the diacetylenicPC at 40 °C disappeared and the bilayers could no longer be photopolymerized. The phase transition and polymerizability of the vesicles could be restored simply by cooling to ca. 10 °C. A similar hysteretic behavior was also observed for pure diacetylenicPC bilayers. Mixtures of 9 and DOPC exhibited phase transitions for both lipids (T = — 18 °C and 39 °C) plus a small peak at intermediate temperatures. Photopolymerization at 20 °C initially proceeded at a similar rate as observed for pure 9 but slowed after 10% conversion. Closed bilayer aggregates, formed from phospholipids (liposomes) or from surfactants (vesicles), represent one of the most sophisticated models of the biological membrane [55-58, 69, 72, 293]. Swelling of thin lipid (or surfactant) films in water results in the formation of onion-like, 1000- to 8000-A-diameter multilamellar vesicles (MLVs). Sonication of MLVs above the temperature at which they are transformed from a gel into a liquid (phase-transition temperature) leads to the formation of fairly uniform, small (300- to 600-A-diameter) unilamellar vesicles (SUVs Fig. 34). Surfactant vesicles can be considered to be spherical bags with diameters of a few hundred A and thickness of about 50 A. Phospholipid vesicles (and bilayers) composed of phospholipids with well-defined fatty acid side chains undergo a sharp transition from a crystallinelike state to an amorphous state as the temperature is raised.107 The transition temperature depends on the nature of the fatty acid side chains. For example, for C12 saturated fatty acid chains on lecithin the transition temperature is 0° and for C18 saturated fatty acid chains it is 58°C for unsaturated lecithins the transition temperature is below zero.107 For real membranes sharp phase transitions are not observed, because of the heterogeneous composition of the membrane.
