On the basis of studies on the metabolism of the fish anesthetic, methane sulfonate of m-aminobenzoic acid ethyl ester (MS-222), by a shark (the spiny dogfish, Squalus aeanthias), Maren et al. (25) reported the formation of a metabolite, m-aminobenzoic acid, by cleavage of the ester bond (Figure 1). This ester-hydrolyzed product was not partitioned across the gill, but was slowly excreted by the kidney. 

Procaine Procaine, the 2-diethylaminoethyl ester of 4-aminobenzoic acid (2.1.1), better known as novocaine, is synthesized in two ways. The first way consists of the direct reaction of the 4-aminobenzoic acid ethyl ester with 2-diethylaminoethanol in the presence of sodium ethoxide. The second way of synthesis is by reacting 4-nitrobenzoic acid with thionyl chloride, which gives the acid chloride (2.1.2), which is then esterified with iV,Ar-diethylaminoethanol. Subsequent reduction of the nitro group by hydrogenation of the resulting ester (2.1.3) into an amino group takes place in the presence of Raney nickel [ 1 ]. 

Studies in Xenopus oocytes are performed with the two microelectrode voltage clamp method. For isolation of the oocytes, the toads are anesthetized using a 1 g/1 solution of 3-aminobenzoic acid ethyl ester and placed on ice. A small incision is made to retrieve sacs of oocytes and is subsequently closed with absorbable surgical suture. On waking up, the toads are placed back into the aquarium. Tricaine methanesulfonate (MS-222). Tricaine methanesulfonate (3-aminobenzoic acid ethyl ester methanesulfonate MS-222) is widely used for the sedation and anesthetization of fish. The compound is 0.01% ionized at body pH and has a partition coefficient of 312. This relatively nonpolar lipophilic drug is rapidly absorbed from water and eliminated in freshwater and marine fish species.

Anesthetize the animals by placing them in a solution of 3-aminobenzoic acid ethyl ester (MS222) (Sigma). A concentration of 1 in 10,000 is required for young larvae, but older animals need a stronger solution, 1 in 1000. The acidic solution should be returned to neutrality with NaOH. Five to 10 min in this solution are usually enough.
