The development of mass spectrometry/mass spectrometry (MS/MS) has provided the chemical analyst with a powerful tool for structure elucidation. The primary goal of this project is to develop the full capacity of triple quadrupole mass spectrometry (TOMS) as a tool for routine structure determination. To accomplish this, we have designed and developed computer data bases for spectra and structures (1,2), programs for matching spectra (3), and procedures... [Pg.321]

Currently, high-performance liquid chromatography (HPLC) combined with atmospheric pressure ionization (API) triple-quadrupole mass spectrometry (MS) is the predominate quantitative technique used in modem pharmaceutical bioanalysis. The key technological achievement in API-MS was the efficient ionization in a liquid stream and transference of ions from atmosphere to vacuum. Of the API approaches developed, electrospray ionization (ESI) is the most commonly used. ESI provides an efficient means of soft ionization amenable to most molecules encountered in a dmg discovery setting. An alternative soft ionization approach is the use of desorption ionization (DI) techniques. The major distinguishing feature of DI techniques is that ions are typically produced from dried samples. [Pg.342]

At about the same time, our laboratory has reported the development and validation of an LC tandem MS assay for as much as six TKIs simultaneously. The proposed LC-MS/MS method allows the simultaneous determination of clinically relevant ranges of concentrations for the six major TKIs currently in use imatinib, dasatinib, nilotinib, sunitinib, sorafenib, and lapatinib [122], Plasma is purified by acetonitrile protein precipitation followed by reversed-phase chromatographic separation. Analyte quantification is performed by electrospray ionization-triple quadrupole mass spectrometry by selected reaction monitoring (SRM) detection using the positive mode. This was the first broad-range LC-MS/MS assay covering the major currently in-use TKIs. [Pg.217]

Triple quadrupole mass spectrometry can provide rapid screening of complex mixtures for specific compounds and can be used to analyze for compounds that cannot routinely be analyzed by GC/MS. In addition, structural information can be obtained for certain types of compounds since in collision-induced dissociations the fragments are likely to show the structual differences of the parent compounds. Complex mixtures have been analyzed by this technique by introducing the sample directly into the heated sample port of the instrument with little or no sample pretreatment. Triple quadrupole mass spectrometry promises to be a useful, cost-effective, and practical advanced technique for environmental analysis, particularly when applied to hazardous waste problems. [Pg.84]

Many instrumental set-ups and geometries have been explored. In triple quadrupole mass spectrometry, the first quadrupole selects the parent ion of interest, the second works as a collision cell to fragment the parent ion, and the third isolates the proper product ion. A hybrid type is the quadrupole time-of-flight (Q-TOF) instrument. [Pg.149]

Tandem mass spectrometers are either triple quadrupole systems (the collision cell is also a quadrupole) or quadrupole ion-trap spectrometers. A triple quadrupole mass spectrometry system is shown in Figure 32F-2. Here, the first quadrupole acts as a mass filter to select the ion of interest. This ion is then fragmented by collision with an inert gas in the collision cell. The final quadrupole mass analyzes the fragments produced. The triple quadrupole system can be operated in other modes. For example, if the first quadrupole is operated as a wide mass filter to transmit a wide range of ions and no collision gas is present in the collision cell, the instrument is operating as an LC/MS system. The instrument can be operated by scanning one or both quadrupoles to produce mass spectra of the fragments of ions selected by the first quadrupole as that quadrupole is scanned. [Pg.981]
