Cells of the T cell lineage appear to be more sensitive to the immunomodulatory effects of Pb compared to other lymphoid populations. In addition, there are considerable differences in sensitivity across various T cell subpopulations [38 41], This differential sensitivity has become another major hallmark of Pb-induced immunotoxicity, although most data implicate T cells as indirect targets of Pb immunotoxicity. Both in vivo and in vitro observations of T-dependent immune responses in the presence of Pb suggest that T helper function and Th-dependent cytokines are skewed preferentially toward Th2 reactivities. Smith and Lawrence [42] found that Pb inhibited antigen presentation and stimulated a T cell clone of the Thl phenotype. McCabe and Lawrence [38] were the first to show that Pb inhibited Thl stimulation while it promoted presentation to Th2 clones. Heo and colleagues [39 41] provided both in vitro and in vivo results supporting this immunomodulation by Pb.

The ability of lymphocytes to recover proliferative function after exposure to authentic N=0 was also assessed. The proliferative response of CTLL-Z, a mouse T cell clone, in response to rmlL-2 24 hr after exposure to authentic N=0 again revealed a 50% diminution of the H-TdR uptake (Fig. 3). The H TdR uptake of a human T lymphocyte clone in response to alloantigen was also inhibited 48 hr after exposure to N=0. Whether the same mechanism responsible for decreased H-TdR uptake of actively proliferating cells (described above) is operative in the system where lymphocytes are exposed to a mitogenic stimulus after -N=0 exposure remains to be determined.

The native calcium-binding protease, calpain, which translocates to the cytosolic surface of membranes upon calcium binding, was enriched from a membrane preparation and shown to elicit high levels of protection (56-67%) in mice (Hota-Mitchell ef a/., 1997). This protein was first implicated in protective immunity as the target of a CD4+ T cell clone that could arm peritoneal macrophages to kill schistosomula in vitro (Jankovic ef a/., 1996). The same clone administered intraperitoneally, conferred 65% protection on irradiated mice challenged via this route with cercariae and recombinant IL-2 (this is a very artificial test from which results should be treated with caution). The use of peptide libraries to study T cell specificity was first demonstrated by Gundlach et al.45 Since then studies have focused on T cell specificity with emphasis on the identification of immunodominant epitopes in infectious diseases and autoimmune disorders, and on the determination of tumor antigens. The PS-SCL approach, as described above, was successfully used to study T cell recognition and to identify and optimize peptides having a range of activities in stimulating proliferative responses, cytokine production, and/or lysis by CD4+ and CD8+ murine and human T cell clones and T cell lines of known and unknown specificity.46-53...
