Streptozotocin diabetes male rats

In streptozotocin diabetic male Wistar rats the activity of total nitric oxide synthase and the amount of mRNAs encoding ecNOS and iNOS depended on the duration of diabetes (Stockklau-ser-Farber et al. 2000). When diabetes had lasted for 4 to 6 weeks, both the total activity as well as the mRNA encoding ecNOS and iNOS were elevated. A decrease of NOS activity and the amounts of mRNA of ecNOS and iNOS was only seen after more than 20 weeks of diabetes. 

Alterations in the capillary filtration of macromolecules are well documented in diabetic patients and experimental diabetes. Various flavonoids, including anthocyanins and ginkgo biloba extracts, have been shown to be effective against experimentally induced capillary hyperfiltration. Cohen-Boulakia et al." demonstrated that anthocyanins were effective in preventing the increase in capillary filtration of albumin and the failure of lymphatic uptake of interstitial albumin in male rats with streptozotocin-induced diabetes. In an earlier study, Valensi and coworkers demonstrated in a placebo-controlled trial that a purified micronized flavonoid fraction (Daflon 500 mg) can improve and even normalize capillary filtration of albumin in diabetic patients.

Diabetic Rats-Phase I. Laboratory rats (CD strain, 250-300g, male) were made diabetic by a single injection of streptozotocin (STZ), 50 mg/kg, into the tail vein. Nondiabetic controls received an equal volume of citrate buffer. Twenty-four hours after the STZ injection, each rat was individually housed for urine collection. The appearance of glucose in the urine (Ames test strips) and a predictable weight loss or depression of the growth curve were taken as confirming evidence of diabetes. 

Male Wistar rats weighing 150-170 g were used throughout the experiment. The animals were maintained on a Purina chow diet and water ad libitum before use. Diabetes was produced by the intravenous injection of streptozotocin at a dose of 50 mg/kg body wt. 

Male Sprague-Dawley rats weighing 150 g to 200 g were used throughout the experiment. Rats were made diabetic by a single intravenous injection of streptozotocin after overnight fasting. 

Human urines (2,0ml) were collected from 5 (22 years old female, 22 yeas old female, 23 years old male 25 years old male and 63 years old male) volunteers. Rat urines (1.0ml) were collected from both normal and streptozotocin induced diabetic rats. For HPLC analysis of GU, Tetra-O-benzoyl derivative of glucose ureide (mp 137-139 C, m/z 639 [M + H+] Mastation JMS-700 in o-nitrobenzylalcohol matrix, Amax 229nm 8 = 8.1037 X 107 Shimazu UV-2001) was employed. Authentic crystal tetra-O-benzyl GU was used as standard.
