Selectivity metabolism

At a more molecular level, the influences of the composition of the membrane domains, which are characteristic of a polarized cell, on diffusion are not specifically defined. These compositional effects include the differential distribution of molecular charges in the membrane domains and between the leaflets of the membrane lipid bilayer (Fig. 3). The membrane domains often have physical differences in surface area, especially in the surface area that is accessible for participation in transport. For example, the surface area in some cells is increased by the presence of membrane folds such as microvilli (see Figs. 2 and 6). The membrane domains also have differences in metabolic selectivity and capacity as well as in active transport due to the asymmetrical distribution of receptors and transporters. 

Based on the idea that pyrrolidines can be metabolized selectively to y-aminobutyric acid derivatives, Wall and Baker [189] developed a retro-metabolic approach with 3-(4-chlorophenyl)pyrrolidine (5.92, Fig. 5.25) as a prodrug for the GABA-ergic agent baclofen (5.93, Fig. 5.25). In vitro studies showed that metabolism of 5.92 yields a pair of isomeric lactam metabolites (5.94, 5.96) and a pair of isomeric amino acid metabolites (5.93, 5.95). Caffeine is completely absorbed within the intestine, and about 97% is metabolized selectively by the liver - relatively independently of perfusion - by way of microsomal demethylation (E. Renner et af, 1984). After intravenous administration of 125 mg caffeine (with 125 mg sodium benzoicum) - equivalent to 1 cups of coffee -a plasma caffeine increase was detected in liver patients, corresponding to the limited demethylation capacity via paraxanthin. The prolongation of half-life and the limitation of plasma clearance (PC) were clearly altered in patients suffering from cirrhosis. The result can be influenced by age, smoking, contraceptives and numerous medicaments, etc. The test shows no side effects, it is cost-favourable and easy to carry out. (56, 70, 73,81,87)... 

This example illustrates the advantage of evolutionary engineering for optimally accommodating a metabolic component into the complex system of cellular metabolism. Selection procedures have also been used to improve more specialized desirable properties such as improved downstream processing characteristics or resistance to phage infection. Although usually undesired, adhesive phenotypes can be selected for the use in certain types of bioreactors that require attachment of cells [108]. 

The anagen or growth phase is characterized by the high metabolic selectivity of the hair matrix and lasts 2-6 years. The maximum length reached by the hair depends on the duration of the eycle. There are large individual differences in this. The metabolism of the hair matrix is wound down over a period of 1-2 weeks in the catagen or transition phase. The dead hair root then remains in the scalp for another 1-6 months in the telogen phase and then falls out (Fig. 2).
