Specificity checks

There are two types of specificity checks that may be warranted when choosing a specific peptide. The first is to demonstrate that the peptide is bound by the desired antibody and not by other, antigenically irrelevant antibodies. An example of this kind of specificity check is shown in Figure 7.3. A peptide that is immunoreactive with the 1D5 estrogen receptor (ER) mAb was covalently bound as a 1 pL spot to the center of each grid location. Sample analysis requires that sufficient test data have been obtained for a sample, such that all internal and external requirements are met. It also covers the accuracy and applicability of test data. Related specific functions include manual and automated result entry, validating against method specification, checking of detection limits, and instrument management. 

The control unit and software. While the assay is being run, the control unit (figure 18.6(6)) displays instructions on an LCD display for the user and directs the flow of data in the instrument. The control unit prompts the user to follow menu-driven instructions to set up the chemistry, run the assay, and collect and store data. A keypad allows the user to respond to the instructions at each step of the assay. As a qualification step, each cartridge undergoes a specification check during the test.

The Robinson Annelation transform has received detailed examination by the LHASA group. One subroutine in the table specifically checks to see if there is any functionality alpha to the ketone in the cyclohexane and if there is, remove it by exchanging it for something non-offensive. This subroutine is reproduced below as an example of the CHMTRN language.

So, calibration is to be performed daily and repeatability (leading to Mmm, see Sect. 29.1.5.2) should be determined regularly as well, for instance weekly. For repeatability also the alternative approach as described in Sect. 29.1.5.2. can be used. All other specific checks can be better performed at a technical service. 

The open framework of Clang seems to be very promising for implementing domain-specific checks (and at the same time it is also a very promising compiler framework). Our mid-term goal is to incorporate some of the features of Stanse and VCC (see Section 4.5) into Clang Analyzer.

From a specification check point of view, then, it is necessary to estimate both FWHM and FWTM. FWFM may be a little difficult to determine because, unless the peak contains a very large number of counts, the channels with counts close to 1/50 of full height will be close to the peak background continuum. Taking the specification in Table 11.3 as an example, the detector would be judged satisfactory relative to the warranted ratios, but not on the ratios given in Table 11.5.
