Collagen-induced arthritis (CIA) in mice is an animal model for rheumatoid arthritis (RA) and can be induced in DBA/1 and C57BL/6 mice using different protocols. The CIA model can be used to unravel mechanisms involved in the development of arthritis and is frequently used to study the effect of new therapeutics. The development of a CIA model in C57BL/6 mice recently enabled researchers to use knockout mice on this background for arthritis research. [Pg.181]

Models of rheumatoid arthritis (RA) were object to l,25(OH)2D3 treatment, providing additional support for the high potential of the hormone for the treatment of Thl-mediated diseases. As an example, in rat adjuvant arthritis l,25(OH)2D3 improved the inhibiting effect of cyclosporine A on arthritis onset as well as on arthritis aggravation [142]. The incidence and severity of type II collagen-induced arthritis was reduced in rats or mice orally treated with 25-(OH)2-D3, the 1,25 (OH)2D3 analog MC 1288 or l,25(OH)2D3 [143-145], l,25(OH)2D3 has further proven to be therapeutically effective in animal models of autoimmune thyroiditis [146], allogeneic transplantation [66, 147,148], rat Heymann nephritis [149] and autoimmune prostatitis [80]. Results from l,25(OH)2D3-related studies in animal models are summarized in Table 10.3. [Pg.339]

In rheumatoid arthritis, a chronic inflammation of joints, the serum and joint fluids contain abnormal complexes, which appear to consist entirely of immunoglobulins. They may be antibody - antiidiotype antibody complexes.Immunization of animals with type II collagen induces a very similar arthri-tis,496,497 collagen probably doesn t supply the... [Pg.952]

Ibrutinib was also active in animal models of several autoinunune disorders such as rheumatoid arthritis and lupus. The racemic version of ibratinib can completely suppress disease progression in a collagen antibody-induced arihriiis model. Ibrutinib itself is also active in arthritis and lupus mouse models by suppressing autoantibody production." ... [Pg.159]

MAPKAP kinase 2 (MK2) plays a key role in the production of pro-inflammatory cytokines such as TNF-a. MK2 is activated by the mitogen-activated protein (MAP) kinase p38 [1-3]. Activated MK2 phosphorylates a number of target proteins in immune cells resulting in cytokine production and cellular proliferation and activation. Mice lacking MK2 are healthy and fertile, but they fail to increase production of pro-inflammatory cytokines such as TNF-a, lL-6, and IFN-y [4] in response to stimuli such as lipopolysaccharide. MK2 knockout mice are resistant to the development of collagen-induced arthritis, a model for human rheumatoid arthritis [5]. The catalytic activity of MK2 is required to mount the pro-inflammatory response [6]. These and related studies have attracted attention to MK2 as a target for the design of therapeutic treatments for rheumatoid arthritis and other TNF-a-driven diseases. [Pg.310]

We have investigated tissue-specific galactosyltransferase abnormalities in an experimental model of rheumatoid arthritis [41], The study focused on whether the observed pathophysiological similarities that develop in both the collagen induced experimental model of arthritis (CIA) and RA are associated with similar glyco-... [Pg.2075]

Pristane-Induced Arthritis (PIA) Model. In one strain of mice, DBA/1, injection of a mineral oil pristane (2,6,10,14-tetramethylpentadecane) into joint regions induces arthritic disease associated with a broad spectrum of autoantibody production, including rheumatoid factor, anti-collagen and antiheat shock protein antibodies, and polyclonal T cell activation (Wooley and Whalen, 1991). Later work showed the ability to modulate the disease by immunoregulatory agents, e.g., administration of IL-12 cytokine (Beech et al., 1997). However, this disease-induction model appears less frequently used, likely due to lower reproducibility between laboratories, and potential... [Pg.185]
