Bioprocess Control An industrial fermenter is a fairly sophisticated device with control of temperature, aeration rate, and perhaps pH, concentration of dissolved oxygen, or some nutrient concentration. There has been a strong trend to automated data collection and analysis. Analog control is stiU very common, but when a computer is available for on-line data collec tion, it makes sense to use it for control as well. More elaborate measurements are performed with research bioreactors, but each new electrode or assay adds more work, additional costs, and potential headaches. Most of the functional relationships in biotechnology are nonlinear, but this may not hinder control when bioprocess operate over a narrow range of conditions. Furthermore, process control is far advanced beyond the days when the main tools for designing control systems were intended for linear systems. [Pg.2148]

Analysis of Phenolic Compounds. A Hewlett-Packard (Palo Alto, CA) Model 1090 HPLC System, was used to determine the levels of specific phenolic components. The HPLC system was equipped with a ternary solvent delivery system, a diode array UV-VIS detector, and HP ChemStation software for data collection and analysis. Full chromatographic traces were collected at 280, 520, 316, and 365 nm, and spectra were collected on peaks. The stationary phase was a Hewlett-Packard LiChrosphere C-18 coliram, 4mm X 250 mm, with 5 pM particle size packing. Operating conditions include an oven temperature of 40 C, injection volume of 25 pL, and flow rate of 0.5 mL/minute. The mefriod was based on a previously published method for phenolic components in wine (30) and used the modified solvent gradient shown in Table II. ABSTRACT Demonstrating compliance with reliability targets in the operational phase is a key requirement in the follow-up of safety instrumented systems. This paper describes a new approach for demonstrating such comphance, taking into accoimt the information from reported failures. This includes to update the failure rate and to make decisions regarding the functional test interval. A brief discussion of some practical experience with data collection and analysis in the operational phase is also included.
