Prostrate

Mode of Motion. The cyclodienes, like lindane and toxaphene, affect the nerve axon produciag hyperactivity, convulsions, prostration, and death. The biochemical lesion is the competitive inhibition of the y-aminobutyric acid (GABA) neurotransmitter binding site of the nerve axon. Spray workers with lengthy exposure to dieldrin have suffered from prolonged and repeated central nervous system disturbances produciag epileptiform coavulsioas. Similar disturbances occurred ia workers heavily exposed to chlordecoae. 

Injected formulation of leuptoleine and triptoreline [57773-63 ] ate used to treat metastasized cancer of the prostrate, whereas an encapsulated formulation of bromocryptin [25614-03-3] is used to inhibit milk production in women after pregnancy. The performance of encapsulated hormonal polypeptides is weU documented (47). Injectable biodegradable mictocapsules loaded with fertUity control agents have been under development for a number of years and have been carried to various stages of clinical development (48) (see Contraceptives). 

By employing C14-labeled pyrethrum, it has been determined that at 35 °C. the rate of penetration of pyrethrum is more than twice that at 15°C. Cockroaches prostrate at 15 °C. can be returned to normal by transferring them to 35 °C., a process which can be repeated for several hours. Since cockroaches transferred from 35° to 15°C. became prostrate more rapidly than those held continuously at 15°C., the insecticide (or some metabolic toxin) was probably in the vicinity of its site of action at 35 °C. but was ineffective. 

Heat prostration—Avoid going outside on hot, sunny days use fans to cool the body if the day is extremely warm sponge the skin with cool water if other cooling measures are not available and wear loose-fitting clothes in warm weather.

Very large doses can cause vomiting, diarrhea, and prostration. Dehydration and congestion occur in most internal organs. Hypertonic solutions can produce violent inflammatory reactions in the gastrointestinal tract.

Effects of Metabolism on Toxicity. Whether the toxic effects seen after exposure to diisopropyl methylphosphonate are caused by the parent compound or its metabolites is unknown. Studies of IMP A show that acute-duration exposure to IMPA results in reduced motor activity, prostration, and ataxia—effects also seen after exposure to diisopropyl methylphosphonate (EPA 1992). Other studies (Little et al. 1986, 1988) show that IMPA, the major metabolite of diisopropyl methylphosphonate, has an affinity for both lung and brain tissues and will bind to proteins in these tissues—effects that were not seen after exposure to diisopropyl methylphosphonate (EPA 1992 Little et al. 1988). These data and other data on the toxicity of IMPA neither support nor contradict the data found in the diisopropyl methylphosphonate studies, so it is not possible to attribute the effects after exposure to diisopropyl methylphosphonate to IMPA. Metabolites of IMPA other than MPA have not been identified. 

Single-dose oral toxicity studies in rats, mice, and cattle indicate that signs of diisopropyl methylphosphonate intoxication include decreased activity, ataxia, tympanitis, and prostration within 1-4 hours after dosing (Hart 1976 Palmer et al. 1979). Mink that received 1,852 mg/kg/day diisopropyl methylphosphonate in their feed displayed aggressive behavior. However, it was concluded that this behavior was probably due to hunger resulting from the unpalatability of the feed (Aulerich et al. 1979).

As discussed in Section 3.1.1, groups of 10 8-w-old Crl CD rats were exposed to aniline vapor/aerosol at concentrations of 359, 400, 453, 530, or 786 ppm for 4 h (E.I.du Pont de Nemours 1982a). No deaths occurred at the lowest concentration. Signs at 359 ppm included cyanosis, tremors, lacrimation, salivation, semi-prostration, an initial body-weight loss followed by normal gain, and a reddish-brown perineal area. 

Signs and Symptoms Characterized by sudden onset of high fever, lethargy, anorexia, stiffness, and incoordination (ataxia). Pulse and respiratory rates are increased. Coughing, diarrhea, dehydration, with frequent urination (pollakiuria) may develop. Prostration and death may occur in a few hours or days. Subclinical cases may be common.
