Prostate cancer proteomic studies

In one study by Hood et al., 282 of 1153 identified proteins were identified by at least 2 unique tryptic peptides from FFPE prostate cancer (PCa) tissue.9 According to the gene ontology classification of the proteins identified, -65% of proteins were predicted to be intracellular proteins, while -50% of the total human proteome is predicted to be located in the intracellular compartment. Additionally, 20% of the proteins identified in the PCa tissue were classified as membrane proteins, which is significantly less than the predicted 40% for the human proteome. This relative disparity is not unexpected, considering the Liquid Tissue sample preparation kit lacks specific protocols for membrane protein extraction. The Liquid Tissue method has also been used for proteomics studies of a variety of FFPE tissue samples, including pancreatic tumors,28 squamous cell carcinoma,4 and oral human papillomavirus lesions.27... [Pg.341]

It is within the context of finding products that have an effect on human health that a study has been published regarding the gene expression of prostate cancer cells treated with pomegranate fruit juice (70) where a comparative proteomics study provided insights for prostate cancer evolution. The involvement of pomegranate fruit products in the molecular mechanism of inducing prostate cancer cell apoptosis has been evidenced. Thus, a possible... [Pg.146]

The use of proteomics in prostate cancer, the most common cancer in men, was reviewed [94]. In vitro studies on prostate cell lines provided positive proof-of-principle results in the identification of novel biomarker when proteomics was utilized to query prostate tissue specimens. 2D-PAGE followed by MS was used to investigate protein profiles in voided urine after prostatic massage in patients with prostate cancer or with benign prostatic hyperplasia [95]. In this study, a potential novel cancer biomarker, calgranulin B/MRP-14, was identified. In another study, agarose 2D-gel... [Pg.121]

The SI LAC approach has also been used to investigate metastatic prostate cancer development at the protein level (Everley et al., 2004). The fact that proteins showed altered concentration ratios by quantitative MS was confirmed by western blotting. In addition, proteomic approaches for quantitation of protein phosphorylation via SILAC combined with MS analysis have been described (Gruhler et al., 2005 Ibarrola et al., 2003, 2004). A recent study reports on identification as well as relative quantitation of in vivo mefhylation sites of proteins in HeLa cells by stable isotope labeling wifh C Hj-methionine (Ong et al., 2004). [Pg.72]
