Propionate Ointment

Melt items 2 and 3 in a fat-melting vessel at temperature 75°C while mixing. 

Start heating mixer vessel to 75°C. Transfer molten items 2 and 3 to mixer through stainless steel mesh under vacuum 0.4-0.6 bar. Start mixer at 10 rpm manual mode. 

In a water bath (temperature 60°C), dissolve item 1 in item 4 using homogenizer for 5 minutes. Add this to mixer with stirring.

Side effects of topical administration of steroids have been reviewed recently (124 ). Further cases with Cushing s syndrome secondary to topical steroid therapy have been reported (131 ). It was shown that treatment with less than 50 g of clobetasol propionate ointment per week has no suppressive effect on the adrenal cortex. When more than SO g of ointment per week was used, a significant number of patients owed adrenocortical suppression (4 ).

Halobetasol propionate is a class I topical corticosteroid, or an ultrapotent corticosteroid according the Stoughton classification system which is widely used in United States. Halobetasol propionate has been approved in the United States for the treatment of corticosteroid-responsive dermatoses. In a number of clinical studies, halobetasol propionate cream and ointment have both been tested against vehicle and compared with other ultrapotent corticosteroids [43]. 

Fluticasone propionate is administered by nasal spray in the prophylaxis and treatment of allergic rhinitis, but is applied topically in the treatment of various skin disorders (as creams and ointments).

Therapeutically, sodium propionate has been used topically in concentrations up to 10% w/w alone or in combination with other propionates, caprylates, or other antifungal agents, in the form of ointments or solutions for the treatment of dermatophyte infections. Eye drops containing 5% w/v sodium propionate have also been used. See Section 18. 

Inappropriate dilution of carefully formulated creams and ointments may result in changes in stability and the effectiveness. The biophar-maceutical considerations will be apparent from the discussion in this chapter. An example would be clobetasol propionate formulated in a base containing the optimum amount of propylene glycol. Stallimycin [inn] (stallimycin hydrochloride [usan] distamycin A) is a (peptide) ANTIBIOTIC active as an ANTIVIRAL. It is used topically to treat herpes simplex infections. Stallimycin hydrochloride stallimycin. Stanolone [ban] (androstanolone [inn] androstanolone propionate androstanolone enanthate dihydrotestosterone) is a steroid, an androgen and ANABOLIC agent. It has been used orally and topically in the form of an eye ointment. 

P-CD, thalidomide by HP-P-CD,539 tauromustine by HP-a-CD,540 and medroxyprogesterone acetate and megestrol acetate by HP-P-CD and methyl-P-CD.541 The stabilizing effect of P-CD against the oxidation of 2-[8-methyl-10,l 1-dihydro-ll-oxodibenz[b,/]oxepin-2-yl]propionic acid in ointments has also been reported.542... 

Transfer the drug mixture from step 3 into the manufacturing vessel from step 2 while mixing. Mix and homogenize for 10 minutes under vacuum at 0.5 bar to obtain uniform homogeneous ointment to contain label amount of fluticasone propionate per gram.
