Fig. 21.4. Vesicle formation and patch-clamp techniques used to record levamisole receptor channel currents from Ascaris muscle. (A) Muscle membrane vesicles bud-off from the bag membrane following a 10 min collagenase treatment and incubation for 1 h at 37°C in Ascaris saline. (B) Levamisole is applied to the outside surface of the membrane to activate receptor channels cell-attached patches are usually used but it is also possible to make inside-out and outside-out patch recordings. Treated rats had 1000 mg/kg FW liver (vs. 4.7 in controls) lowered hemoglobin, hematocrit, and red cell counts mean survival time of 67 days hepatic and renal histopathology Dose-time-dependent increase in copper concentrations in liver, spleen, and lung little accumulation in muscle and skin. Reduced growth at 2.5 and 3.75 mg/kg BW daily reduced survival at 3.75 mg/kg BW. Maximum copper concentrations recorded, in mg/kg FW (vs. saline controls,) were 710 in liver (<5), 212 in kidney (<10), 7 in lung (<1.5), 27 in spleen (<2.0) 6 in bone (<2.0) and 2.2 in testes (<1.6) Increased serum ceruloplasmin and white blood cell number... [Pg.206]

Partition coefficients of a series of aliphatic hydrocarbons, including n-hexane, have been determined in human tissues (Perbellini et al. 1985). The following partition coefficients for n-hexane (olive oil/air, blood/air, tissue/air) were determined olive oil, 146 blood, 0.80 liver, 5.2 kidney, 3 brain, 5 fat, 104 muscle, 5 heart, 2.8 and lung, 1. Saline/air partition was not reported separately for n-hexane, but was very low for the range reported for the entire group of compounds (0.1-0.4). [Pg.95]

The cholinesterase-inhibiting activity of the phosphorofluoridates was compared quantitatively with that of eserine sulphate thus. To 0-2 ml. of heparinized human plasma was added 05 ml. of a solution containing either eserine or the phosphorofluoridate in varying concentrations then the mixture was kept at room temperature for 10 min. before 1 /tg. of acetylcholine in 1 c.c. saline solution was added. After 5 min. at room temperature, the mixture was made up to 10 ml. with frog saline containing eserine 1/100,000, which at once stopped the action of any cholinesterase not yet inactivated. The solution was then assayed for acetylcholine on the frog rectus-muscle preparation. [Pg.75]

Figure 6. Semi quantitative evaluation of spontaneous adsorption of collagen IV conjugated with a fluorescence marker Oregon Green 488 (A) to polyethylene foils iradiated with ions (energy 30 keV, dose from lO to lO ions/cm ), and its correlation with the number of rat aortic smooth muscle cells initially adhering to the polymer on day 1 after seeding (B). Collagen was diluted in phosphate-buffered saline to the concentration of 0.02 mg/ml (10 pg/cm ) and incubated with the foils for 24 h at room temperature. Mean+SEM from 4-9 experiments. Student t-test for unpaired data [43]. Figure 6. Effects of WSAP on Guinea Pig Ileum. In preliminary experiments with a constant infusion of 5 pg/m1 of WSAP in the physiological saline bath the following were observed A) a slow tonic contraction of the smooth muscle segment resulting in a shift of the base line of approximately 30%, B) a latency period of approximately 10 to 15 minutes before there was any significant... [Pg.261]

At the sites of Injection of oxime in the rabbits, various extents of hemorrhage and of purulent, indurative myositis and muscle necrosis were seen. The rabbits that received physiologic saline and five that received intramuscular injections of 2.15 M sodium chloride on 8 d in a satellite experiment had waxy degeneration of muscle at the site of injection. Five rabbits that received intramuscular injections of 2.15 M I on 8 d also had waxy degeneration of muscle at the site of injection. This was stated to be more extensive than that seen in the rabbits given equimolar sodium chloride. No other lesions in the rabbits that seem to be attributable to the oximes were described. [Pg.273]

In each of these methods, undiluted serum, urine, acid-deproteinized milk, or a buffered saline extract of muscle was mixed with sulfamethazine-horseradish peroxidase and added to antibody-coated wells of a microtiter plate. A sulfamethazine-bovine serum albumin conjugate prepared by the glutaraldehyde procedure (56) was used for antibody production. Results showed that screening of serum was of value since sulfamethazine concentrations in serum directly correlated with those in swine tissues. Thus, for example, a level of 100 ppb of sulfamethazine in... [Pg.843]

Fig. 13.1 Cytoprotective action of adenosine in a quantitative model of mouse hindlimb ischemia and reperfusion (I/R) injury. Adult wild type mice were injected with (a) sterile vehicle (0.1% DMSO in phosphate-buffered saline, pH 7.4) or various adenosine receptor agonists (b, c) or antagonist (d, f). (a) Following ischemia and reperfusion, skeletal muscle showed a significant uptake of Evans Blue dye (EBD) in vehicle-treated mice (first part of (a), representative of 7 mice). The contra-lateral leg not subjected to ischemia-reperfusion showed virtually no EBD uptake. In the second part of (a), the same section was stained with rabbit polyclonal anti-skeletal muscle actin antibodies followed by staining with goat anti-rabbit IgG conjugated with FITC. (b) The nonselective adenosine agonist R-PIA caused a large reduction in the EBD-stained area (see both first and second parts to this figure, representative of six mice)...
