However, recombinant antibodies may be less stable and have lower binding affinities than monoclonal antibodies (Valle and Jendoubi, 2003). Therefore, in order to fully implement the microarray format, a host of diverse capture agents could be required in addition to antibodies. These include peptides, small molecules, aptamers, ribozymes, or other molecular recognition probes yet to be discovered. However, it is also xmderstandable because of the diverse nature of proteins that additional technologies besides microarrays will be used in proteomics research (Hanash, 2003). 

Diluted human serum (1 pL) was incubated with the peptide microarray and bound antibodies were detected using a rhodamine-labeled anti-human IgG. Signal was detected using a slide scanner (Affymetrix model 418) with data collection in the Cy3 channel. A reported eightfold gain in sensitivify at 100% specificity over standard ELISA was achieved using the peptide microarray.

The role that carbohydrates exert in recognition phenomena of biological and pharmaceutical relevance has stimulated the quest for tools that can monitor and even quantitatively evaluate these interactions. The potential diagnostic applications of such tools is evident, and the possible development of commercial carbohydrate microarrays, such as those established for oligonucleotides, and very recently peptides, is of high interest.

Oligonucleotide and peptide microarrays can be prepared in situ with light-directed synthesis on a glass surface in conjunction with either a photolithographic method or using a micromirror device (2,11-15). However, these methods are not useful for most organic synthesis. Furthermore, such approaches require equipment that is not readily available. Spot synthesis on cellulose membrane is another in situ synthesis method, but the resulting microarrays are low density (16).
