Inadvertent isolation

Low Level Waste. The NRC 10CFR61 specifies the nature of the protection required for waste containers (20). Class A wastes must meet minimum standards, including no use of cardboard, wastes must be solidified, have less than 1% Hquid, and not be combustible, corrosive, or explosive. Class B wastes must meet the minimum standards but also have stabiHty, ie, these must retain size and shape under soil weight, and not be influenced by moisture or radiation. Class C wastes must be isolated from a potential inadvertent intmder, ie, one who uses unrestricted land for a home or farm. Institutional control of a disposal faciHty for 100 years after closure is requited. 

Opening a manual valve. Manual valves which are normally closed to isolate two or more pieces of equipment or process streams can be inadvertently opened, causing the release of a high-pressure stream or resulting in vacuum conditions. Other effects may include the development of critical flows, flashing of liquids, or the generation of a runaway chemical reaction. 

Water flowed from the relief pipe (20 ft elevation) to tank E610. It first flowed through the jumper to the process pipe to the normally closed blow-down valve which was open (possibly inadvertently left open or failed to seat in the attempt to pressurize the tank). About 119 gallons of water flowed through this open blow-down valve, through another, normally open, isolation valve to enter tank E610, where it reacted with MIC. 

While contractors were working in a building, they inadvertently burned some insulation material. The ventilation system spread the fumes around the building. Two people were affected by them, and an expensive experiment taking place in a laboratory was ruined [15]. Before authorizing hot work in a building, consider the effects of any fumes that might be produced and, if necessary, switch off or isolate the ventilation system. 

On the basis of the information contained in a step-by-step procedure, a checklist can be designed consisting of active checks for critical steps which operate as an "aide memoir." Experienced workers who normally carry out a task from memory may forget isolated acts or inadvertently not follow the correct sequence of steps in the procedure. 

A minor explosion in the use of gaseous acetyl hypofluorite was caused by inadvertent contact of the gas inlet tube with a cooling bath, which condensed liquid hypofluorite which subsequently exploded on contact with organic material or air [1], More detail on the isolation of this compound of limited stability, particularly in the liquid phase, is given [2],... 

To avoid inadvertent degradation of this kind during isolation and fractionation of the starch, an oxygen-free atmosphere appears essential. Perhaps the use of an oxidation inhibitor (for example, hydroquinone, quinol, or V-phenyl-2-naphthylamine) would also be suitable. 

The variations in the value of molecular weight may be due to the poly-molecularity of the samples and to inadvertent degradation during isolation. 

In resting neutrophils, about 50% of the total cellular FcyRIII pool is expressed on the cell surface. There is considerable variation in this value because many methods used to isolate neutrophils can also inadvertently mobilise these subcellular receptors. The remainder of the total cellular FcyRIII that is not expressed on the plasma membrane is present in the subcellular pool. However, if the FcyRIII normally present on the plasma membrane is cleaved (e.g. via the action of elastase or pronase) and the cells subsequently activated, then FcyRIII reappears on the cell surface via the mobilisation of these pools. Thus, the expression can be restored to up to 70% of the resting level within 15 min via such a translocation. During activation (and presumably priming), FcyRIII (together with other plasma membrane markers) is also translocated to the plasma membrane however, because the receptor is also shed from the cell, the total number of receptors on the cell surface remains largely unchanged. There is also some evidence that continued expression of FcyRIII on the cell surface requires de novo biosynthesis of this receptor (see Fig. 7.8). 

Many nitrate salts of amines are highly crystalline, which encourages their use to isolate amines they are also often explosive, which does not. An industrial explosion consequent upon inadvertent isolation of such in part of a production plant, with subsequent overheating is reported [1]. 

In some cases, when petroleum and/or petroleum products are released to the environment, a free phase is formed and sample(s) of the hydrocarbon material can be collected directly for characterization. The ability to analyze free product greatly aids the determination of product type and potential source. The samples may be diluted prior to analysis EPA SW-846 3580 (waste dilution) gives some guidelines for proper dilution techniques. However, caution is advised since as part of the initial sample collection procedure, water and sediment may be included in the sample inadvertently. Several protocols involved in initial isolation and cleanup of the sample must be recognized. In fact, considerable importance attaches to the presence of water or sediment in crude oil (ASTM D1796, D4007), for they lead to difficulties in other analyses. 

For sorbitol and mannitol the parameters were from Ref. 2 while those for MTHP are described in Table V below. Our parameters correspond to a set devised to be used with the explicit addition of water if water is to be considered. While we cannot exclude the possibility that some information from solution has inadvertently been included in those parameters, we believe that the isolated molecule will be reproduced if solvent is not explicitly present. 

Sustained acid hyperscretion, due either (usually) to a duodenal or pancreatic gastrinoma, or to multiple endocrine adenomata, responds well to proton pump inhibition, often requiring high doses. Resection should always be considered as a cure for those with solitary adenomata, as should operation to reverse the retained antrum syndrome (partial gastrectomy where the antrum is inadvertently retained and isolated from acid inhibition, and secretes gastrin continuously). 

Once a recombinant gene is constructed and introduced in bacteria or yeast, the expressed protein may provide greater purity and quality, through tight production control, than one can obtain from an extracted protein. Added benefits of switching from a isolated natural protein to a recombinant form are reduced variation of biologic activity and less inadvertent contamination.
