At the sectoral level, national sector-specific CP strategies should be developed. This would allow CP interventions at company level to be put into a broader perspective and would point out possible synergies between companies, suppliers and customers, allowing gains obtained at the sector and national levels to be optimised. It would also enable adequate choices in technology development and transfer in the country as well as the preparation of coherent investment plans to be presented to potentially interested companies and financial institutions (Fig. 1).

This book describes the principal physico-chemical techniques for characterising the catalysts used in searching for new active phases, optimising the formulation and monitoring industrial production. Based on courses given at the Institut Fran ais du Petrole for research technicians in the fields of kinetics and catalysis, this book covers useful basic theory and provides numerous examples of industrial applications. 

This chapter describes the early hit discovery project to identify inhibitors of Hsp90 at Vemalis in 2002-2003. This generated the initial ideas from which series of preclinical candidate compounds were discovered, some of which continue in clinical trials for treatment of cancer [11, 17]. The very early work (virtual and fragment screening) was carried out within Vemalis before a collaboration was initiated with the group of Paul Workman at the Institute of Cancer Research. Subsequently, the project was partnered with Novartis, who were responsible for preclinical development and clinical trials while work at Vemalis continued on identification and optimisation of backup and follow-up compounds.

A highly useful source for finding novel lead compounds constitute the compound collections of synthetic small-molecular-weight compounds that have been accumulated over the past decades in industrial companies and academic institutions. Since these compound collections are usually derived from a limited amount of so-called priviledged core structures such as benzodiazepines, dihydropyrimidines, phenothiazines and others, they exhibit a more limited chemical and structural diversity than natural products. Due to their ususally simpler structure a given hit or lead can be subsequently optimised in a quite efficient manner resulting in low manufacturing and development costs.

Of course I realise every craftsman has to test his tools, but in opening this important NATO Advanced Studies Institute and Summer School I want to focus for a few minutes on the role and general methodology of our subject, I have two reasons. The first is to optimise our own work and maximise our contribution to research. Secondly I am concerned that the subject of computational physics has such a low status. Most physicists consider computation as an occupation for failed theorists and failed experimentalists. This attitude, which has all sorts of consequences for funding and jobs, will only change if we articulate clearly the role of computational physics. And with the capabilities of modern machines, I believe we are at the beginning of an enormous and exciting development so that there is a lot to say. 

Nuclear power was put into an existing technical and institutional infrastructure without much changing this infrastructure - still characterized by the use of oil in particular but also of coal and gas [i.e., nuclear was deployed in an energy supply architecture optimised for fossil].
