This chapter describes the early hit discovery project to identify inhibitors of Hsp90 at Vemalis in 2002-2003. This generated the initial ideas from which series of preclinical candidate compounds were discovered, some of which continue in clinical trials for treatment of cancer [11, 17]. The very early work (virtual and fragment screening) was carried out within Vemalis before a collaboration was initiated with the group of Paul Workman at the Institute of Cancer Research. A highly useful source for finding novel lead compounds constitute the compound collections of synthetic small-molecular-weight compounds that have been accumulated over the past decades in industrial companies and academic institutions. Since these compound collections are usually derived from a limited amount of so-called priviledged core structures such as benzodiazepines, dihydropyrimidines, phenothiazines and others, they exhibit a more limited chemical and structural diversity than natural products. Due to their ususally simpler structure a given hit or lead can be subsequently optimised in a quite efficient manner resulting in low manufacturing and development costs. [Pg.22]

Of course I realise every craftsman has to test his tools, but in opening this important NATO Advanced Studies Institute and Summer School I want to focus for a few minutes on the role and general methodology of our subject, I have two reasons. The first is to optimise our own work and maximise our contribution to research. Secondly I am concerned that the subject of computational physics has such a low status.
