Open healing is slower than when the re-epithelializing wound is covered. Test or control material is applied either onto or under an occlusive dressing to the abraded or unabraded shaved skin of animals. Wound healing can be assessed by applying large (for example 1 g/1 kg) topical doses of test material to an epithelial wound (for example, an incision) and monitoring wound healing over a period of 14 days.

Nutritional vitamin A deficiency causes xerophthalmia, a progressive disease characterized by night blindness, xerosis (dryness), and keratomalacia (comeal thinning), which may lead to perforation xerophthalmia may be reversed with vitamin A therapy. However, rapid, irreversible blindness ensues once the cornea perforates. Vitamin A also is involved in epithelial differentiation and may have some role in corneal epithelial wound healing. There is no evidence to support using topical vitamin A for keratoconjunctivitis sicca in the absence of a nutritional deficiency.

Change the apple juice agar plates every 1-2 h and age appropriately to obtain embryos at the required developmental stage. Stage 15 embryos should be used for epithelial wound and hemocyte wound assays, which requires aging embryo collections overnight at 18°C. 

Timelapse image epithelial wound closure by confocal microscopy. The Z-stack should be large enough to observe the entirety of the epithelium which will be approximately 2-3 pm thick. One minute between images is sufficient to quantify wound closure. 

Timelapse imaging of epithelial wound closure should be watched and subde focal adjustments down into the sample will be required as the epithelium is sealed shut.

According to their physiological conditions, wounds can be classified into five types, each with a symbolic color code necrotic wounds, sloughy wounds, granulating wounds, epithelializing wounds, and infected wounds, which are characterized by their black, yellow, red, pink, and green appearances respectively. Figure 7.1 illustrates the five types of wounds.
