To evaluate the separate fractions two methods are available. Vertical dividing lines can be dropped from each trough of the curve to the base line, but this ignores the overlapping of the fractions (which varies since not all the fractions are equally widely separated). Initially this technique was intended purely as a high-resolution imaging device, down to the sub-nanometre level (Alessandrini and Facci, 2005). Calibration is necessary for in-situ spectrometry in TLC. Either the peak height or the peak area data are measured, and used for calculation. Although the nonlinear calibration curve with an external standard method is used, however, it shows only a small deviation from linearity at small concentrations [94.95 and fulfils the requirement of routine pharmaceutical analysis 96,97J. One problem may be the saturation function of the calibration curve. Several linearisation equations have been constructed, which serve to calculate the point of determination on the basis of the calibration line and these linearisation equations are used in the software of some scanners. A more general problem is the saturation function of the calibration curve. It is a characteristic of a wide variety of adsorption-type phenomena, such as the Langmuir and the Michaelis-Menten law for enzyme kinetics as detailed in the literature [98. Multidetector GPC software , a PC-DOS package written by Lesec (6), was used for triple detection GPC. The major limitation to using CTA-SI to estimate the infarct core is the requirement that image acquisition occurs during a steady-state concentration of intravascular contrast. With the advent of faster, multidetector CT scanners, CTA imaging likely occurs during the upslope of the concentration-time curve and thus violates the steady-state assumption.
