A large proportion of this immediate increase in sales is due to nonloyal buyers (brand switchers). An HPLC method for chlorogenic acids with lactones in six different commercial brands of roasted coffee was developed by Schrader et al. (143). Hydroxycinnamic acid derivatives, including mono- and di-caffeoylquinic acids, corresponding lactones, and feruloylquinic acids were extracted from coffee with methanol at 80°C for 1 h under reflux. An HPLC method using step-gradient elution with 2% aqueous acetic acid (eluent A) and ACN (eluent B) for a 75-min run time was developed. Determination was carried out by HPLC with UV detection at 324 nm, and further confirmation was conducted by HPLC-thermospray (TSP)-MS and HPLC-diode array detection. Elution order for mono-caffeoylquinic acid (CQA) was 3-CQA, 5-CQA, followed by 4-CQA, which was different from the usual elution order of mono-CQA (Fig. 17). These results indicate that it is currently not possible to predict the elution order of different reversed-phase packings due to the different selectivity (143). [Pg.814]

The truth is that coffee is a strong stimulant, one that is hard on certain parts of the body. It is irritating to the stomach, for example, and many people who drink a lot of it have indigestion most of the time. (In the United States, where coffee is regularly consumed in large quantities, there are nearly as many brands of antacids as there are brands of coffee.) It is irritating to the bladder, too, especially in women, and is a frequent cause of urinary complaints. Note that decaffeinated coffee is not inert. In addition to small amounts of caffeine (enough to stimulate sensitive individuals), it contains other active substances from the coffee bean that can be irritating to the nervous, gastrointestinal, cardiovascular, and urinary systems. Even when speed began to get a bad reputation in the late l960s I didn t know I was on it. I was really shocked to learn that Desoxyn was lust a brand name for methamphetamine and even more shocked to find that I couldn t stop taking it. If I tried to cut down the dose or stop, I would be groggy, depressed, and unable to concentrate or do my school work. I would also eat a lot and gam weight. The main ingredient in OTC stimulants is caffeine. Plienylpropanolamine (Pl A) was a common ingredient in such preparations until 2000, when tlie FDA withdrew OTC approval because it was found to increase the risk of stroke. Popular brands of OTC stimulants are No-Doz and Vivarin. They certainly will induce mild central nervous system stimulation, but No-Doz, for example, contains about as much caffeine as one or two cups of coffee, so the user should expect about that effect. To extract the ephedrine from the pills, the first thing which must be done is to grind them into a fine powder. This pulverization must be thorough in order to ensure complete extraction of the ephedrine from the filler matrix in which it is bound. While an occasional cup of coffee, aspirin or No-Doz will keep you awake, and is relatively harmless, pay special attention to seeing that you don t take in too much caffeine for too long a time.
