Uracil is used more effectively, in nucleic acid synthesis within a rat hepatoma than in normal liver. This observation appears to have stimulated the synthesis of 5-fluorouracil (1027) as an antimetabolite mainly because the introduction of a fluorine atom involves a minimal increase in size. In the event, 5-fluorouracil did prove to have antineoplastic activity and it is now a valuable drug for treatment of tumors of the breast, colon or rectum, and to a lesser extent, gastric, hepatic, pancreatic, uterine, ovarian and bladder carcinomas. As with other drugs which interfere with DNA synthesis, the therapeutic index is quite low and great care is required during treatment (69MI21301). [Pg.152]

It is used by direct instillation into the bladder for multifocal local bladder carcinoma. Nausea and myelosuppression are the major toxicities of thiotepa. It is not a local vesicant and has been safely injected intramuscularly and even intra-thecally. [Pg.56]

Most kidney transplant procedures are heterotopic, where the kidney is implanted above the pelvic bone and attached to the patient s iliac artery and vein. The ureter of the transplant kidney is attached directly to the recipient s bladder or native ureter. The native kidneys usually are not removed, and data have shown that under most circumstances, removal of the native kidneys does not influence patient survival and allograft survival. Special circumstances, however, such as renal cell carcinoma and PCKD, may necessitate native kidney removal.1,3 There were 16,477 (9914 deceased donors, 6563 living donors) kidney transplants and 903 simultaneous kidney-pancreas procedures done in 2005.3... [Pg.831]

A 2-year carcinogenicity bioassay of tricresyl phosphate in mice and rats showed no evidence of carcinogenicity in these species (NTP 1994). Doses (consumed in feed) were <37 mg/kg/day in mice and < 15 mg/kg/day in rats. Dietary administration of tributyl phosphate was associated with transitional and squamous cell carcinomas of the bladder in rats after 2 years of exposure at 143.3 mg/kg/day (FMC 1994a). An increased incidence of hepatocellular adenomas in the liver was observed in mice after dietary administration of 455 mg/kg/day tributyl phosphate for 18 months (FMC 1994b). [Pg.131]
