The mechanism of the blood pressure-lowering activity of the /3-blockers, which have been used as antihypertensives for several decades on a very large scale, is still not understood in full detail. Aniracetam (6), launched in 1993 in both Japan and Italy for the treatment of cognition disorders, is in Phase II trials in the United States as of this writing. In clinical studies it has been shown to cause some improvement in elderly patients with mild to moderate mental deterioration (63), and in geriatric patients with cerebral insufficiency (64). In a multicenter double-blind placebo-controUed trial involving 109 patients with probable AD, positive effects were observed in 36% of patients after six months of treatment (65), a result repeated in a separate study of 115 patients (66). Powder metallurgy is used in the fabrication of fuel elements as well as control, shielding, moderator, and other components of nuclear-power reactors (63) (see Nuclearreactors). Treatment of Manic—Depressive Illness. Siace the 1960s, lithium carbonate [10377-37-4] and other lithium salts have represented the standard treatment of mild-to-moderate manic-depressive disorders (175). It is effective ia about 60—80% of all acute manic episodes within one to three weeks of adrninistration. Lithium ions can reduce the frequency of manic or depressive episodes ia bipolar patients providing a mood-stabilising effect. Patients ate maintained on low, stabilising doses of lithium salts indefinitely as a prophylaxis. However, the therapeutic iadex is low, thus requiring monitoring of semm concentration. Adverse effects iaclude tremor, diarrhea, problems with eyes (adaptation to darkness), hypothyroidism, and cardiac problems (bradycardia—tachycardia syndrome). [Pg.233]

Other agents are also used for the treatment of manic-depressive disorders based on preliminary clinical results (177). The antiepileptic carbamazepine [298-46-4] has been reported in some clinical studies to be therapeutically beneficial in mild-to-moderate manic depression. Carbamazepine treatment is used especially in bipolar patients intolerant to lithium or nonresponders. A majority of Hthium-resistant, rapidly cycling manic-depressive patients were reported in one study to improve on carbamazepine (178). Carbamazepine blocks noradrenaline reuptake and inhibits noradrenaline exocytosis. The main adverse events are those found commonly with antiepileptics, ie, vigilance problems, nystagmus, ataxia, and anemia, in addition to nausea, diarrhea, or constipation. Carbamazepine can be used in combination with lithium. Several clinical studies report that the calcium channel blocker verapamil [52-53-9] registered for angina pectoris and supraventricular arrhythmias, may also be effective in the treatment of acute mania. Its use as a mood stabilizer may be unrelated to its calcium-blocking properties. Verapamil also decreases the activity of several neurotransmitters. Severe manic depression is often treated with antipsychotics or benzodiazepine anxiolytics. [Pg.233]

A number of studies have shown that vitamins moderate the induction of chromosomal aberrations by radiation. Vitamins C and E given orally to mice either 2 h before, immediately after, or 2 h after 1 Gy (100 rad) of y-ray TBI significantly reduce the frequencies of micronuclei and chromosomal aberrations in BM cells. Vitamin E is the more effective (95). Administration of vitamins C and E within 5 min of irradiation is as effective as pretreatment. Protection by vitamin C has also been shown in humans. Whereas chronic treatment of rats using vitamin C (100 or 300 mg/(kg/d)) for six months prior to TBI protects against chromosomal aberrations, vitamin E is not radioprotective in this setting (96). [Pg.491]

Pet Foods and Commercial Animal Feeds. Eor many years, it has been known that stable, long-shelf-life, intermediate-moisture pet foods can be prepared through the use of 0.1—0.3 wt % sorbates. In these products, the antimicrobial effectiveness of sorbates is enhanced by a combination of moderate heat treatment, pH adjustment, and reduced water activity via humectants such as propylene glycol, or by adjusting sugar and salt content. Precipitation softening processes are used to reduce raw water hardness, alkalinity, siHca, and other constituents. This helps prepare water for direct use as cooling tower makeup or as a first-stage treatment followed by ion exchange for boiler makeup or process use. The water is treated with lime or a combination of lime and soda ash (carbonate ion). These chemicals react with the hardness and natural alkalinity in the water to form insoluble compounds. The compounds precipitate and are removed from the water by sedimentation and, usually, filtration. Waters with moderate to high hardness and alkalinity concentrations (150—500 ppm as CaCO ) are often treated in this fashion. [Pg.259]
