Thaumatin extraction

Thaumatin. Thaumatin [53850-34-3] is a mixture of proteins extracted from the fmit of a West African plant, Thaumatococcus daniellii (Beimett) Benth. Work at Unilever showed that the aqueous extract contains two principal proteins thaumatin I and thaumatin II. Thaumatin I, mol wt 22,209, contains 207 amino acids in a single chain that is cross-linked with eight disulfide bridges. Thaumatin II has the same number of amino acids, but there are five sequence differences. Production of thaumatins via genetic engineering technology has been reported (99). [Pg.281]

The extraction, purification, or concentration of samples for thaumatin determination will depend on the type of food matrix, as described in Sec. I.C. Purification can also be accomplished by partition (126). Ramsohoye and Kozlov (124) purified thaumatin on a cation-exchange cartridge (CM Sephadex C-50). After conditioning the cartridge with phosphate buffer, the sample is loaded and washed with buffer. Thaumatins are then eluted with 0.25 M sodium chloride. Mackenzie et al. (127) separated thaumatins on cellulose CM52. After equilibration of the column with buffer, pH 7.2, the sample was applied, washed with the buffer, and eluted with a gradient of 0 to 0.2 M NaCl in the same buffer. [Pg.546]

Proteins were then dissolved in 20 mL of 0.125 M Tris-HCl, pH 7.4 containing 1% (m/v) CHAPS and the protease inhibitor cocktail, and treated with CPLL. With several variations from the standard treatments such as other complementary extractions from seeds, the use of urea-containing extraction buffer, and an additional peptide ligand library, the total number of detected gene products was quite large. Two hundred and thirty one unique proteins were found in the pulp (vs. 56 described previously) and 61 in the seeds (vs. only four reported by the literature). In the latter case, the presence of seed storage proteins, oleosins, and histones were detected in the pulp, a thaumatin-like protein, an allergenic protein also named Ole el3, was confirmed. [Pg.143]

This protein sweetener is extracted from the berry of a fruit which grows in west Africa. It has been sold under the trade name Talin. Thaumatin is intensely sweet. The sweetness is slow in onset but is long lasting with a liquorice end-note. It is little used in confectionery although it has been used as a coating on the outside of chewing gum where its persistence would be an advantage. [Pg.137]
