Thaumatin-like proteins provide a sweet taste in food. They may display antifungal activity and are treated as potential allergens. These proteins are found in strawberries, apples, bell peppers, and cherries (Scheurer et al., 1999 Aalberse et al., 2001). [Pg.117]

Proteins were then dissolved in 20 mL of 0.125 M Tris-HCl, pH 7.4 containing 1% (m/v) CHAPS and the protease inhibitor cocktail, and treated with CPLL. With several variations from the standard treatments such as other complementary extractions from seeds, the use of urea-containing extraction buffer, and an additional peptide ligand library, the total number of detected gene products was quite large. Two hundred and thirty one unique proteins were found in the pulp (vs. 56 described previously) and 61 in the seeds (vs. only four reported by the literature). In the latter case, the presence of seed storage proteins, oleosins, and histones were detected in the pulp, a thaumatin-like protein, an allergenic protein also named Ole el3, was confirmed. [Pg.143]

In all cultivars of V. vinifera studied so far, thaumatin-like proteins and chitinases are the major soluble components of grapes (Peng et al. 1997 Pocock et al. 1998,2000 Tattersall et al. 1997). The predominance of these PR proteins was clear at all stages of berry development following vdraison (Pocock et al. 2000). Importantly, as the concentration of these proteins in the berries continually increases... [Pg.216]

The other major class of proteins of the grape berry are the Thaumatin-like proteins (TLP) (Tattersall et al., 1997). In addition to the main TLP isoform called VVTL1, another minor TLP form can be detected (Pocock et al., 2000). These proteins show, respectively, an apparent MW of 24 and 25 kDa by SDS-PAGE, but their MW, determined by electrospray ionization-mass spectrometry, (ESI-MS) is 21.272 and 21.260kDa (Pocock et al., 2000). Moreover, the MW of the TLP identified by the proteomic analysis of the whole Cabernet sauvignon... [Pg.252]

Grape proteins have been separated on C18 (Santoro, 1995 Peng etal., 1997) and C8 (Tattersall et al., 1997) stationary phases. A widely used column is the Vydac C18 (Peng et al., 1997 Waters et al., 1998), which allows separation of different forms of grape thaumatin-like proteins (WTL) and chitinases (Chit) (Figure 7.8). [Pg.270]
