Early 2005, Leadbeater s team reported that the previously claimed tran-sition-metal-free Suzuki-type protocol was definitely palladium-catalyzed [ 53 ]. Palladium contaminants down to the level of 50 ppb found in commercially available sodium carbonate were responsible for the generation of the biaryl. For good product yields in a short reaction time under microwave irradiation, a loading of 1 ppm Pd was required. 

In a related publication, Kobayashi and his team reported on Zr-catalyzed asymmetric Mannich reactions that utilize the more electron-rich oxygenated ketene acetals shown in Scheme 6.28 [93], A noteworthy aspect of this study was that the levels of syn/anti diaste-reocontrol proved to be dependent on the nature of the alkoxide substituent whereas the (3-TBS acetals predominantly afforded the syn isomer, the OBn derivatives afforded a larger amount of the anti isomer. As before, the presence of an additive, this time 1,2-dimeth-ylimidazole (DMI), proved to be important with regard to the level of Ti-facial selectivity. The phenol activating group can be removed by the same procedure as reported previously, with essentially identical degrees of efficiency (see Scheme 6.27). 

As mentioned above, much less new information on imidazo[5,l-c][l,2,4]triazines appeared during the past years. Only one Russian team reported synthetic pathways to this ring system, and the results are summarized in Scheme 53. 

An Italian team reported that pyridazinones (89) hydroxymethylated at C-5 induced a high decrease in systolic blood pressure in rats [371]. In Italy, much effort has been devoted also to the preparation of conformationally restricted congeners of antihypertensive pyridazinones. In a structure-activity study, it has been found that indeno[l,2-c]pyridazinones, in particular compounds (90), are potent antihypertensive agents [372]. 

Information derived from data-gathering activities serves as the basis for valid conclusions and recommendations. Without effective data gathering, the incident cannot be defined or analyzed effectively. In some cases, gathering data can consume most of the time and resources spent by the investigation team. Some teams report that it can take up to 70 percent of the investigation effort depending on the nature of the occurrence. 

Their proposal won FDA approval, the first gene therapy trial everto inject AAV. In June 1999 the clinical team injected the first patient with genetically altered AAV. The procedure was uneventful. Two more patients followed. Hamilton and coworkers designed a rocket propellant formulation based on BAMO-AMMO Copolymer TPE and 80% total solids (AP and Al) and it was directly cast into a phenolic case and required no liner or insulation. The results of firing of a 18 kg test motor exhibited completely acceptable ballistic properties. The authors claimed it to be the 1st demonstration of a high energy TPE motor firing [158]. Prior to this, Wardle and his team reported BAMO-AMMO copolymer and CL-20-based potentially attractive high energy ETPE gun propellants. 

Management should verbally respond to the inspection findings during the discussion of the FDA-483. Each item should be discussed individually, and the company personnel should provide additional explanations where appropriate and should state their intentions for items where they have made or intend to make improvements. When companies have initiated corrective actions, it is imperative that the FDA be informed of the actions taken (especially corrections that have already been completed). The company should request that the FDA team report in their EIR the corrections that have been accomplished. If... 

The balance of participants time is spent studying the Dowville case and preparing team reports to be delivered at the end of the workshop. In the reports, the team identifies the root causes of the Dowville incident, identifies deficiencies in Dowville s S/LP/S programs that allowed the root causes to exist, and recommend actions to correct those deficiencies. The team presentations are judged by a panel of managers with one report selected as the winner.

The same team reported recently on a similar reaction of 282 aiming at a one-pot preparation of heteroaryl amide 285 <2004TL5909>. However, in this case, the malononitrile was successfully used as a carbonyl synthon instead of A /V-diethylaminoacetonitrile <2004TL5909>. 

Porco s team reported the formation of benzopyrylium salts 29 (Scheme 5.14) from the action of silver and gold catalysts on o-alkynylbenzaldehydes 28.54 Although this reaction proceeds much faster with gold than with silver, the benzopyrylium formation is of interest and will be discussed further. 

The joint Dubna-Berkeley research team reported the discovery of ununpentium in 2003. Of the 30 atoms thus far observed, the one with the longest half life is ununpentium-288, with a half life of about 100 ms. 

In 2006, the Dubna-Berkeley research team reported that they had detected either three or four atoms of ununoctium, with a half life of about 0.89 ms (which is in doubt because of the limited number of atoms studied). 

In 1973, Semerdzieva and Nemd first reported the existence and qualitative content of psilocybin in collections of Psilocybe bohemica. This research team reported psilocybin levels of up to 1.1 % in dried samples. Results from my own studies also revealed variable levels of alkaloids in different mushrooms collected at the one location near Sazava.

That independence is critical. An in-house quality assurance (QA) person may qualify as long as reporting relations establish complete independence from the facility management.
