The moist crude bromohexadiene is quaternized in water without further purification. The submitters report that, if desired, the crude mixture may be dried over anhydrous calcium chloride and fractionally distilled through a 10-in. stainless-steel-packed column at reduced pressure. SFC chromatographs represent hybrids between GC and HPLC instruments (Fig. 6.4). In order to deliver the supercritical fluid, syringe pumps or reciprocal pumps are used and maintained above the critical temperature using a cryostat regulated at around 0 "C. In instances where an organic modifier is used, a tandem pump is employed which has two chambers, one for the critical fluid and one for the modifier. The liquid then passes through a coil maintained above the critical temperature so that it is converted into a supercritical fluid. Stainless steel packed columns like those used in HPLC (1 to 4 mm in diameter) or fused silica capillary columns like those used in capillary GC (2 to 20 m in length, internal diameters as low as 50 pm and stationary phase film thickness of at least 1 pm) are used in SFC. [Pg.98]

Smith et al (76,773 analyzed hydralazine in a drug mixture containing hydralazine hydrochloride, hydrochlorothiazide, and an impurity derived from the latter. The column was 1 m x 2.1 mm (ID) stainless steel, packed with a strong anion exchanger on 30 Jm Zipax . A preparative column of dc = 50 mm (50 000 p,m) packed with 15 p,m particles contains approximately 3000 particles over the column diameter. For column stability the column wall plays only a minor role and the packing depends on the mechanical stability of the bed. The column is made of carbon steel, but both packings are of stainless steel. The column will operate under vacuum with conditions of 55 kPa and bO C at the top and 60 kPa and 125°C at the bottom. Using molecular sieve 13X it is possible to separate paraffinic and naphthenic hydrocarbons according to carbon number (22]. However, the retention of high boiling hydrocarbons was very high. For example, the time required for the elution of hydrocarbons up to Cii in a typical naphtha sample on a packed column is 1.5 h (see Fig. 2-3a [21]).
