Matheson et al. (1978) reported the results of a battery of in vivo and in vitro assays to assess the genotoxicity of 1,1-dimethylhydrazine. Included were the Ames Salmonella microsome assay, a microbial suspension assay, mutation induction at the TK locus in L5178Y mouse lymphoma cells, stimulation of UDS in WI-38 cells, and a dominant lethal assay in mice. 1,1-Dimethylhydrazine was active in all of the tests except the dominant lethal assay. [Pg.189]

In Chapter 1 Bruce N. Ames, the plenary speaker at the symposium, describes the development of the Salmonella/microsome assay, which is better known as the Ames Test. This test was the first proven rehable and rapid in-vitro method for the mutagenic and carcinogenic screening of chemicals. Included is an excellent overview of the potential problem of human exposure to toxic substances as well as a description of how this important method is being used to test chemical substances. [Pg.296]

Genotoxicity studies (Salmonella microsome assays, sister chromatid exchange, and tests of unscheduled DNA synthesis and of chromosome aberrations in cultured mammalian cells) indicate that selenite, selenate, and selenide have both genotoxic and antigenotoxic effects (Biswas et al. 1997, 2000 Gairola and Chow 1982 Khalil 1994 Lu et al. 1995b Schillaci et al. 1982 Ueda et al. 1997 van der Lelie et al. [Pg.210]

Genotoxic Effects. There are some data on in vitro genotoxicity of carbon disulfide. Haworth et al. (1983) evaluated carbon disulfide at five doses both with and without S9 activation in the Salmonella/microsome assay. All results were negative. Other studies in Salmonella typhimurium and Escherichia coli, both with and without activation, were also negative (Donner et al. 1981 Hedenstedt et al. 1979). Salmonella strain TA98 was used in a host-mediated assay investigating the ability of carbon... [Pg.96]
