DESYMMETRIZATION AND ENZYMATIC STRATEGIES FOR CHIRAL PROPARGYL ALCOHOL SYNTHESIS... [Pg.155]

Aconitatc was obtained as a minor product in the carbonylation of propar-gyl alcohol[479]. However, in the two-step synthesis of methyl aconitate (536) from propargyl alcohol in 70% overall yield, the first step is the oxidative carbonylation under CO and air using Pdli and KI to giNe dimethyl hydro-xymethylbutenedioate (535), which is carbonylated further to give trimethyl aconitate (536) by u.sc of [Pd(Tu)4jl2 as a catalyst[480]. [Pg.99]

Synthesis of trisubstituted olefins via LAH reduction of the propargylic alcohols and iodination as the key step (Ref. 1). [Pg.146]

The synthesis of enantiomerically pure propargylic alcohols is possible using the same methodology 43b. Thus, addition of (—)-[(l-chloro-2-phenylethyl)sulfinyl]-4-methylbenzene (14) to propan-al led to a mixture of the diastereomers 15A/15B (d.r. 44 56) which are easily separated by column chromatography. After thermal elimination of the sulfinyl group the vinyl chlorides 16A/16B were obtained as a mixture of E- and Z-oleftns. Elimination of hydrogen chloride was carried out with three equivalents of butyllithium, leading to enantiomerically pure 1 -phenyl-1-pentyn-3-ol. [Pg.138]

Enyne metathesis starting either from acetylenic boronates and homoallylic alcohols [104a,c] or from propargyl alcohols and allylboronates [104b] has recently been described. The resulting boronated dienes can be converted to allenes or cycloaddition products. The cross metathesis of vinylcyclopropyl-boronates directed toward the total synthesis of natural products has very recently been investigated by Pietruszka et al. [104d]. [Pg.256]

Friedel-Crafts reaction remains unexplored, possibly due to the difficulty of the cycloalkyne formation. A mild, versatile, and efficient method for the one-step synthesis of substituted dihydro- and tetrahydroisoquinolines has been developed by the FeCl3-6H20-catalyzed intramolecular allenylation/cyclization reaction of benzylamino-substituted propargylic alcohols, representing the first example of the intramolecular Friedel-Crafts reaction of propargylic alcohols (Scheme 8) [24, 25]. FeCls, InCls, and Yb(OTf)3 also exhibit good catalytic activity for the reaction. [Pg.7]

We now report the synthesis of new antibacterial 3H-pyrazoles by regioselective 1,3-dipolar cycloaddition of the versatile 2-diazopropane to nonprotected disubstituted propargyl alcohols and that the unsubstituted propar-gyl alcohol allows the double addition of 2-diazopropane and gives a 3H-pyrazole with formal insertion of the dimethylcarbene into a carbon-carbon bond. We also show that the photolysis of the 3H-pyrazoles leads to new alcohols containing the cyclopropenyl unit. [Pg.144]

In 2002, Braga el al. employed a chiral C2-symmetric oxazolidine disulfide as a ligand for the enantioselective synthesis of propargylic alcohols by direct addition of alkynes to aldehydes (Scheme 3.64). Good yields but moderate enantioselectivities (<58% ee) were obtained for the enantioselective alkyny-lation of aldehydes in the presence of ZnEt2. [Pg.144]

Later, a nickel-catalyzed cascade conversion of propargyl halides and propargyl alcohol into a pyrone in water was reported. The reaction involved a carbonylation by CO and a cyanation by KCN (Eq. 4.55).96 Recently, Gabriele et al. explored a facile synthesis of maleic acids by palladium catalyzed-oxidative carbonylation of terminal alkynes in aqueous DME (1,2-dimethoxyethane) (Eq. 4.56).97... [Pg.127]

The final example demonstrates that microwave irradiation allows a perfect fine-tuning of reaction conditions to obtain different products from the same starting materials. In the procedure developed by Garcfa-Tellado and coworkers [41], two domino processes were coupled. The first process consists of a high-yielding synthesis of enol-protected propargylic alcohols 10-111 starting from alkyne 10-109 and aldehyde 10-110 (Scheme 10.28). In the second process, transformation into... [Pg.583]

Schubert, T., Hummel, W., Kula, M.-R. and Muller, M. (2001) Enantioselective synthesis of both enantiomers of various propargylic alcohols by use of two oxidoreductases. European Journal of Organic Chemistry, (22), 4181—4187. [Pg.164]

Tejedor and coworkers have utilized a combination of two domino processes for a microwave-promoted synthesis of tetrasubstituted pyrroles [344]. The protocol combines two coupled domino processes the triethylamine-catalyzed synthesis of enol-protected propargylic alcohols and their sequential transformation into pyrroles through a spontaneous rearrangement from 1,3-oxazolidines (Scheme 6.183). Overall, these two linked and coupled domino processes build up two carbon-carbon bonds, two carbon-nitrogen bonds, and an aromatic ring in a regioselective and efficient manner. The tetrasubstituted pyrroles could be directly synthesized from the enol-protected propargylic alcohols and the primary amines by microwave irradia-... [Pg.225]

Azido derivatives of furazans have proved to be particularly useful for the synthesis of heterocyclic systems. Thus, by 1,3-dipolar addition of l-azido(4-amino-l,2,5-oxadiazol-3-yl)aldoxime 186 to propargyl alcohol and phenylacetyl-ene, bicyclic 4-amino-l,2,5-oxadiazol-3-yl(4-R-l,2,3-triazol-l-yl)ketoximes 187 were obtained (Equation 34) which in reaction with acetic anhydride afforded the corresponding 0-acyl derivatives <2003RJ0574>. [Pg.351]

Palladium-catalyzed cyclization reactions with aryl halides have been used to synthesize pyrazole derivatives. V-Aryl-lV-(c>-bromobenzyl)hydrazines 26 participated in a palladium-catalyzed intramolecular amination reaction to give 2-aryl-2W-indazoles 27 . Palladium-catalyzed cascade intermolecular queuing-cyclocondensation reaction of o-iodophenol (28) with dimethylallene and aryl hydrazines provided pyrazolyl chromanones 29 <00TL7129>. A novel one-pot synthesis of 3,5-disubstituted-2-pyrazolines 32 has been achieved with an unexpected coupling-isomerization sequence of haloarene 30, propargyl alcohol 31, and methylhydrazine <00ACIE1253>. [Pg.169]

Muller and co-workers reported the three-component one-pot synthesis of various pyrimidines through the in situ generation of unsaturated carbonyl compounds. The palladium catalyzed coupling of aryl halides bearing electron withdrawing substituents 7 with propargyl alcohols 8 produced unsaturated carbonyl compounds 9 after isomerization, which condensed with amidines 10 to form triaryl pyrimidines 11 . [Pg.262]

Various transition metal complexes, in particular of late transition metals, were reported to be effective catalysts for such double bond isomerization. Because organic synthesis is the focus of this volume, this section will cover the transition metal-catalyzed isomerization of alkenes, which has the significant synthetic and industrial utilities. This chapter will also include the synthetic application, asymmetric reactions,4-6 and isomerization of alkynes, in particular, that of propargylic alcohols. [Pg.71]

A ruthenium(n)-indenyl complex, which is an efficient catalyst for the isomerization of allylic alcohols, is also an effective catalyst for the isomerization of propargylic alcohols to both a,/3-enals and a,/ -enones (Scheme 57).96 In this reaction, the addition of 20—40 mol% InClj is highly effective. The reaction exhibits extraordinary chemoselectivity and a variety of functional groups are unaffected, which allows a highly efficient synthesis of dienals (R1 =Me2C = CH, R2 = H). [Pg.97]

This ruthenium-catalyzed isomerization of propargylic alcohols was successfully applied to the synthesis of a,(3-cnal 6, a key intermediate in the total synthesis of sphingofungin analogs (Equation (30)).97... [Pg.97]

The acyclic version of Larock s heteroannulation was successfully applied to the synthesis of highly substituted pyridines [166]. The annulation of rert-butylimine 210 with phenyl propargyl alcohol produced pyridine 211 regioselectively in excellent yield. The regiochemistry obtained was governed by steric effects. Furthermore, the choice of imines was crucial to the success of the heteroannulations. terr-Butylimine was the substrate of choice, since all other imines including methyl, isopropyl, allyl and benzyl imines failed completely to produce the desired heterocyclic products. [Pg.225]

To make tryptophan analogs, Gronowitz and coworkers conducted a pyrrole annulation from an aminoiodopyrimidine utilizing the Larock indole synthesis conditions (see Section 1.10.) [80]. They prepared heterocondensed pyrrole 115 by treating 4-amino-5-iodopyrimidine 114 with trimethylsilyl propargyl alcohol under the influence of a palladium catalyst The regiochemical outcome was governed by steric effects. [Pg.398]
