Overlayer method

Optimization of the RP HPLC system is critical for maximum sample throughput compatible with the mass spectrometer data acquisition capabilities. For highest throughput, the samples should be injected into the HPLC column or directly into the mass spectrometer using the overlay method, i.e., during the ESI-MS assay of a sample, the next sample is retrieved and prepared for injection immediately... [Pg.83]

There are several ways to coat a MALDI plate. One is two-layer overlayer [62,63] In this method, the first layer on the MALDI target plate comprises the densely packed matrix microcrystals formed by the quick solvent evaporation of a matrix solution in acetone or a mixture of acetonitrile and water at room temperature. A mixture solution containing both matrix and sample is then deposited onto the first layer of small crystals to form uniform analyte-matrix microcrystals. The overlayer method is reported to provide improved results, particularly for analyses of proteins and peptides [64-66],... [Pg.403]

Biocompatibility at the Cell Culture Level. The liquid components, MAP and catechol oxidase, and extracts of two solid crosslinked materials were tested for cell culture toxicity and growth inhibition. The evaluation of cell culture cytotoxicity in an agar overlay method detects the impact on cells of any freely... [Pg.471]

Comparison of the X-ray based cubic section model with the substrate surrogate derived cubic section model gave a view of the amino acid residues around the substrate binding domain (Figure 21). Forbidden and boundary regions defined by the substrate surrogate overlay method are near the side chains amino acid residues. Substrate interactions with surface of the amino acid residues could be estimated by molecular mechanics (MACROMODEL). [Pg.540]

The evaluation involved four basic methods and variants of them. They included the standard visual overlay method, or a modification of it that involved some linear measurements in the fingerprint region of the infrared spectrum the Brown log-ratio method (26) the vector method devised by Killeen et al. (44,45) and the standard ASTM method (22). The overlay method was the best method when used by someone experienced in interpreting weathered oils. [Pg.72]

Batista et al. (58) used an overlay method in the bioassay-guided fractionation of an acetone extract of the roots of Plectranthus hereroensis (Labiatae) to isolate the antibacterial diterpene (Compound 14). Staphylococcus aureus was used as the test organism. [Pg.241]

Because predictions are made for each unit area, the overlay method is strong in predicting spatial patterns, although weak in estimating magnitudes a rather elaborate set of rules is often required to reveal differences in severity of impacts from place to place. [Pg.32]

The objectivity of the overlay method is high with respect to the spatial positioning of effects and impacts, but is otherwise low. Overlays are not effective in estimating or displaying uncertainty and interactions. [Pg.32]

The overlay method provides guidance on the spatial design of inspection procedures to be followed. [Pg.33]

Overlay techniques have a long history of use in environmental planning being ideally suited for consideration of spatial aspects. They rely on a set of maps of environmental characteristics (physical, social, ecological and aesthetic) that are overlaid to produce a composite characterisation of the local environment. A modem version of the overlay method is the computer-based geographical information system (GIS). In simple terms, a GIS stores, retrieves, manipulates and displays environmental data in a spatial format. A set of maps or overlays of a given area provide different types of information and scales of resolution. The main drawbacks are the lack of appropriate data and the expense of creating a usable system. [Pg.156]

The multicellular spheroids were initiated by the agar overlay method described by Yuhas. After 6 days in culture, spheroids with diameters between 200 pm and 300 pm were selected for growth experiments and transferred to 16mm multiwell culture dishes. The diameters of the spheroids were determined by using an inverted microscope with a calibrated graticule in the eyepiece. The wells were base-coated with 0.5ml 0.75% DMEM-agar and filled with 1 ml of medium. [Pg.206]

GIS-type comparisons can be undertaken, enabling planners to study a few more alternatives than they would ever do if it were to be done manually by the overlay method. This finally should result in an overall better quality of planning. [Pg.411]

The signal overlay method is not considered to be very sensitive, and is highly dependent on the resolution of... [Pg.613]

Overlayer (also Two Layer and Seed Layer) Method this method (several variants have been developed by different research groups) is a combination of the crushed crystal and fast evaporation methods. Fast solvent evaporation is used to form the first layer of small matrix-only crystals, followed by deposition of a solution of matrix plus analyte on top of the crystal layer. The difference between the fast evaporation and the overlayer methods is in the second layer solution, where the addition of matrix to the latter is believed to provide improved results particularly for mixtures. The method has several convenient features that make it a popular approach. It inherits all the advantages of the fast evaporation method and avoids some of its limitations, while providing enhanced sensitivity and better spot-to-spot reproducibility. [Pg.243]

Sandwich method this method is derived from the fast evaporation and overlayer methods. The sample analyte is not premixed with matrix instead a sample droplet is applied on top of a fast evaporated matrix-only bed as in the fast evaporation method, followed by deposition of a second layer of matrix in a nonvolatile solvent so that the sample is sandwiched between the two matrix layers. This method is not widely used. [Pg.244]

In XPS a method of determining the attenuation length is to use the overlayer method. The variation of the intensity of a species is determined as a function of the thickness of the overlayer. If the substrate is a silicon wafer and the overlayer has a thickness d, then the intensity of the silicon signal will be 7s, which according to eqn (9.39) is given by... [Pg.260]

BSA was not included in the renaturation buffer nor was the membrane blocked with 5% BSA or other proteins as described elsewhere (Ferrell and Martin, 1991 Celenza and Carlson, 1991) because this reduced the signal observed from CaM kinase II. Incubation of the membrane with BSA or other proteins may, however, increase the signal from other kinases by providing a substrate. Other procedures have employed incorporation of microtubule-associated protein 2 (MAP-2) into the gel matrix to augment detection of CaM kinase II (Kameshita and Fujisawa, 1989) using the gel overlay method however, MAP-2 is a substrate for multiple kinases, thus reducing the specificity of the assay. [Pg.262]
