Not all tumors are seriously harmful. The body is able to wall off some tumors so that they do not spread such noninvasive tumors are said to be benign. Other tumors, called malignant, invade the body and destroy normal body tissues and organs. In advanced stages of cancer, malignant tumors may develop the ability to spread to other parts of the body and initiate new tumors, a process called metastasis. [Pg.164]

In humans, also, preferential sites exist for the formation of metastasis from various primary tumors [reviewed in Zetter (Zl)]. Thus, bone is a preferred site for metastasis from primary malignancies in breast, prostate, and kidney, while liver is a frequent metastatic site for tumors originating in the colon. Different types of leukemias vary widely in their ability to spread to liver, lymph, bone, and spleen. Some organs, however, are rarely colonized by metastatic growth. These resistant sites include skeletal muscle, heart, and skin. [Pg.138]

Ewing s hypothesis may explain certain patterns of metastasis observed in humans, such as the colonization of regional lymph nodes from some primary cancer in humans. There are, however, human cancers that form distant metastases in a variety of organs without any evidence of local lymph node involvement. [Pg.139]

It now appears that both mechanical factors and finding a suitable environment are responsible for the preferred organ location of secondary growths. The relative importance of these factors is unknown and may vary depending on the type of tumor. Proctor (P4) has suggested that the initial dissemination of neoplastic cells is controlled mostly by mechanical factors, whereas later metastasis is determined more by appropriate environment. [Pg.139]

Benign tnmonrs are nsnally only a problem when they impair the fnnction of organs or canse metabolic stress. They can be removed by snrgery or radiation therapy. Malignant tnmonrs are mnch more of a problem, since the cells can escape from the primary tnmonr to other sites in the body, where they settle and develop into secondary tumours (metastases). In another rat study, 3,3 -diehlorobenzidine was administered to 50 male (70 mg/kg/day) and 50 female (80 mg/kg/day) Sprague-Dawley rats, in a standard diet for up to 16 months (Stula et al. 1975). In rats fed 3,3 -diehlorobenzidine in the diet for a total of 349 days (females) and 353 days (males), histopathologieal evaluations revealed mammary adenocarcinoma (16% incidence), malignant lymphoma (14%), granuloeytie leukemia (20%), carcinoma of the Zymbal gland (18%) in males, and mammary adenocareinoma (59%) in females. These tumors were either totally absent or occurred statistically less frequently in untreated controls. The authors noted that most of these tumors appeared to arise in the bone marrow and hematopoietic foci in the spleen and liver with subsequent metastasis to other organs. Only one dose level was used in the study, however, and information on the prrrity of the test substance was not provided. [Pg.49]
