APase in onion roots. Enzyme activity was mainly extracellular with the heaviest concentration in corner spaces between the epidermal and hypo-dermal layers. He suggested the possibility of a subcutaneous pore through which the enzyme could be released to the root surface. Bieleski and co-workers (Reid Bieleski, 1970 Bieleski Johnson, 1972) studied the psi induction and location of APase in duckweed (Spirodela oligorrhiza). APase in control plants was located primarily in and around the vascular strands. In P-deficient plants psi-APase activity was 10-20 times the control value. Enzyme activity was primarily located in the epidermis of the root and undersurface of the frond, the tissue locations most likely to provide access to phosphate esters in the medium. These workers further demonstrated that hydrolysis of organic phosphates occurred in the external medium and/or the apoplast followed by Pi uptake into the cell. [Pg.28]

Stiles et al.43 found artemisinin to inhibit respiration in Lemna minor, but the compound strongly stimulates oxygen uptake by lettuce root tips.6 All mitotic phases of onion root tips are inhibited by the compound, and it induces a low level of abnormal mitotic figures.6 Dayan et al.6 conducted a battery of simple physiological tests with several artemisinin analogues. Holotoxin Ai inhibits the RNA biosynthesis in Candida albicans and Saccharomyces carlsbergensis, as indicated by the decrease in incorporation of C-uridine to the acid-insoluble fraction of the cells. Similar results were obtained for glycoside fractions of 14 species of Pacific sea cucumbers [132]. Apparently the inhibition of RNA biosynthesis in Saccharomyces carlsbergensis is related to nucleotide loss from yeast cells after treatment with glycosides. The action of these active substances on plant cells has been studied by Richmond and Phillips (1975) using onion (Allium cepa) as test object. Benomyl and carbendazim induce identical aberrations in the cells of onion root. Benomyl acts considerably more rapidly than carbendazim.
