Offline methods

A variety of online solid extraction devices and applications have been developed for bioanalysis. Many are easy to build in laboratories or commercially available. Unlike offline methods, minimal operator intervention is needed for daily sample analysis after online applications are set up, so the approach is both labor- and cost-effective. The technique can also minimize errors arising from manual operations, eliminate potential inconsistencies caused by different operators, and provide accessibility of LC/MS/MS applications to laboratories that have minimal analytical expertise. [Pg.280]

Step 5 Off-line method or analyzer development and validation This step is simply standard analytical chemistry method development. For an analyzer that is to be used off-line, the method development work is generally done in an R D or analytical lab and then the analyzer is moved to where it will be used (QA/QC lab, at-line manufacturing lab, etc.). For an analyzer that is to be used on-line, it may be possible to calibrate the analyzer off-line in a lab, or pseudo-on-line in a lab reactor or a semi-works unit, and then move the analyzer to its on-line process location. Often, however, the on-line analyzer will need to be calibrated (or re-calibrated) once it is in place (see Step 7). Offline method development and validation generally includes method development and optimization, identification of appropriate check samples, method validation, and written documentation. Again, the form of the documentation (often called the method or the procedure ) is company-specific, but it typically includes principles behind the method, equipment needed, safety precautions, procedure steps, and validation results (method accuracy, precision, etc.). It is also useful to document here what approaches did not work, for the benefit of future workers. [Pg.395]

The proposed strategies for stabilization of gas-lifted oil wells are offline methods which are unable to track online dynamic changes of the system. Online IS introduction allows loading of samples in the biological matrix without preparation. ISs were introduced online in the quantitation of propranolol and diclofenac in plasma (Alnouti et al. 2006). Plasma samples were loaded into the autosampler without pretreatment. Both the plasma sample (10 /iL) and IS (5 //I. from an IS microreservoir) were aspirated into an injection needle sequentially and injected into the sample loop. After the switching of an injection valve, the mixed solution in the sample loop was loaded into a cartridge containing washing solution for online SPE. Online SPE LC/MS/MS is commonly used for bioanalytical applications in the pharmaceutical industry. Column switching systems and TFC systems are easy to build and control. Sophisticated commercial systems and SPE cartridges are readily available. Compared to offline sample preparation, the online approach can save time and labor. However, the development of online SPE bioanalytical assays remains analyte-dependent. Generic methods can be applied to many analytes. For extremely hydrophobic, hydrophilic, and ionic analytes at normal pH range and analytes with a variety of hydrophobicity and pKa values, analyte-specific methods must be developed. An understanding of the chemistry of the analytes and SPE is critical. [Pg.293]

Inclusion of a second pump for effluent dilution and transfer supports both online and offline MS analysis.
