A large number of proto-oncogenes code for transcription factors required for progression of the cell cycle and/or for the differentiation of the cell. The best known and investigated examples of oncogenic mutated transcription factors involve the jun, fos and myc genes and the genes for the T3 receptor and the vitamin A acid receptor. 

The c-myc gene is presented as an example of overexpression of a transcription factor due to translocation.

The c-myc gene, identified originally as the cellular homolog of the transforming determinant carried by avian myelocytomatosis virus, is altered in association with a broad spectrum of neoplasms. Consistent with the observation that altered c-myc is associated with tumors of diverse origin, it has been observed that normal c-myc is expressed in a variety of tissues. Thus, c-myc appears to encode a function associated with a ubiquitous metabolic pathway. 

The c-myc gene encodes a 49-kd protein that is highly concentrated in the nucleus of the cell. The concentration of c-myc protein normally varies appreciably with the metabolic state of the cell, increasing by more than an order of magnitude in cells just prior to chromosome duplication and cell division. 

In most human tumors associated with the c-myc gene, the concentration of c-myc protein is greatly amplified. Burkin s lymphoma, which we discussed earlier, is a notable exception. In this case the c-myc transcript is marginally, and in some cases not at all, increased by comparison... 

Members of the myc gene family have been implicated in the control of normal cell proliferation as well as in neoplasia. A more direct role for myc genes in transformation is indicated by their ability to transform primary rat embryo fibroblasts in association with the c-H-ras oncogene.
