It was found that the HIV envelope glycoprotein in vitro increases the production of NO by human monocyte-derived macrophages [114]. NO production is increased in patients who have AIDS [115], and the increased concentrations of nitrite in AIDS patients with opportunistic infections is caused by T gondii, Pneumocystis carinii, Mycobacterium tuberculosis, and Mycobacterium avium, whereas nitrite concentrations are normal in symptom-free patients. It was also confirmed that there was increased production of NO in the sera of children with HIV-1 infection, and of circulating cytokines, such as interleukin lp, tumor necrosis factor a, and interferon y. It is postulated that rises in the concentrations of these cytokines may represent a substantial stimulation of NO production [116]. In contrast, it has been shown that there was no altered endogenous nitrate formation in eight patients with AIDS, most of whom had opportunistic infections [117]. It has also been noted that there were high... [Pg.20]

The j8-VLDL receptor has been identified on mouse peritoneal macrophages [13,14], foam cells associated with rabbit aortic atherosclerotic plaques [58], and human monocyte-derived macrophages [14]. Although )8-VLDL can bind to LDL receptors, LDL do not compete with I-labeled jS-VLDL for uptake by mouse peritoneal macrophages [13,14], In addition, monocytes from LDL receptor-negative FH patients or from normal subjects express equivalent numbers of j8-VLDL receptors [14]. The j8-VLDL receptors thus are distinct from LDL receptors. The -VLDL receptor is also distinct from the receptors for chemically modified lipoproteins (see below). [Pg.49]

Phagocytosis of apoptotic cells by macrophages leads to the production of anti-inflammatory cytokines as a way of preventing inflammation. Kurosaka et al. [79] have demonstrated that human serum potentiates the production of two anti-inflammatory cytokines, IL-10 and TGF-J3, by both TPA-treated THP-1 cells and human monocyte-derived macrophages. This enhanced response to the presence of apoptotic cells also results in the suppression of the production of the pro-inflammatory cytokine IL-8. In addition, human IgG and FcyRI appear to be critical in triggering the production of anti-inflammatory cytokines by macrophages in response to apoptotic cells. [Pg.158]
