Clinical Investigations Diesel exhaust (mass median diameter <10 pm) at 300 pg/m associated with concentrations of NOj of 1.6 ppm, NO of 4.5 ppm for 1 h, CO of 7.5 ppm, total hydrocarbons of 4.3 ppm, formaldehyde of 0.26 mg/m and 4.3 x 10 suspended particles/ml in healthy human volunteers exposed for 1 h induced a 240 % median relative increase in interleukin-8 mRNA gene transcripts in the cells contained in bronchial lavage performed 6 h later (Salvi et al. 2000). There were no changes in the mRNA transcript levels for ILL-ip, ILL-4, TNF-a, interferon-y, and granulocyte-macrophage colony-stimulating factor after exposure to diesel exhaust compared with air. [Pg.355]

The functional modulation of the effect of cytokines requires the presence of cellular and soluble receptors. The expression of these receptors is regulated by specific signals, often dependent on cytokine networks. Transiently produced and of short half-life, cytokines are typically not stored as preformed molecules, and their production is dependent on transient genetic transcriptional activation and short-lived messenger ribonucleic acid (mRNA). Cytokines are rapidly secreted, leading to a brief burst of cytokine release. This burst is very important for cytokines to achieve distant activities, contrasting with the small amount of these molecules necessary for local functions (see later section on interleukin-1). [Pg.646]

TNFa and the interferons decrease lipogenesis, by downregulating the mRNA levels of the key lipogenic enzymes acetyl-CoA carboxylase and fatty acid synthase the interferons diminish mRNA levels of fatty acid synthase but not of acetyl-CoA carboxylase. TNF-a, interleukin-1, and some interferons also increase lipolysis, although the mechanism is probably post-transcriptional, since Northern blot analysis shows that TNF-a and the interferons decrease the level of perilipin mRNA. [Pg.301]

Expression of vascular cell adhesion molecule-1 (VCAM-l) and intercellular adhesion molecule-1 (ICAM-1) is known to be elevated at sites of inflammation. Studies have been conducted into the effects of EGCG and TF-3 on the expression of these adhesion molecules induced by interleukin-1(3 (IL-ip) in cultured human umbilical vein endothelial cells (HUVECs) [20]. Both compounds significantly inhibited IL-ip-induced protein expression of VCAM and ICAM in dose-depen-dent manners and were associated with reduced adhesion of leukocytes to HUVECs. The mRNA level of VCAM-l was also inhibited by these tea polyphenolics, as was the NF-xB-dependent transcriptional activity induced by IL-ip. It is concluded that these molecules exhibited... [Pg.197]

The RNA dot hybridization with human IL-6 cDNA showed that panaxatriol saponin enhanced the interleukine-6 mRNA concentration within PHA activated peripheral blood mononuclear cell by about four folds, suggesting that panaxatiol saponin could promote the IL-6 gene transcription of lymphoid tissue[49]. [Pg.72]
