Solution 1 is prepared from exactly 5 ml of 0.4092% w/v atropine sulphate solution and exactly 1 ml of 2.134% w/v homatropine hydrobromide solution. The solution is basified and extracted, the solvent is removed and the residue is treated with 2 ml of BSA and then diluted to 50 ml with ethyl acetate. Solution 3 is prepared from exactly 2 ml of eyedrops and exactly 1 ml of 2.134% w/v homatropine hydrobromide solution. Homatropinae Hydrobromidum Homatropine Hydrobromide, C16H21N03HBr, is the hydrobromide of an alkaloid obtained by the condensation of tropine and mandelic acid.

It is soluble in water (1 6) and alcohol (1 40), less soluble in chloroform (1 420). and insoluble in ether. 

Homatropine hydrobromide is used topically to paralyze the ciliary structure of the eye (cyclopicgia) and to effect mydriasis. It behaves very much like atropine but is weaker and less toxic. In the eye. it acts more rapidly but less persistently than atropine. Dilation of the pupil takes place in about IS to 20 minutes, and the action subsides in about 24 hours. By using a miotic, such as physostigminc, it is possible to restore the pupil to normality in a few hours.

Wyatt et al. worked out a GLC assay for atropine and scopolamine in belladonna extract. The extract was solved in 0.1 N sulphuric acid, homatropine hydrobromide was added to this solution as an internal standard, and interfering materials were extracted from the acidified solution with chloroform - and finally a mixture of chloroform and 2-propanol (10 3) if there is an emulsion problem.
