Synthesis of the vaccine candidate 21 was achieved in high yield by condensation of the C-terminal carboxylic acid of lipopolypeptide (24) and the amine of spacer-containing Tn antigen (25), followed by deprotection of the amino acid side chain. The fully protected lipopolypeptide 24 was produced by coupling the lipid S-[2,3-bis(hexadecanoyloxy)-propyl]-7V-Fmoc-Cys (Pam2FmocCys (3)) to the N-terminal amine of the resin-bound peptide (22), and by subsequent linking of the hexadecanoic add to the free amine of the resin-bound lipopolypeptide (Fig. 14).

The role of a hydroxylation step is consistent with the effects of iron chelators and anti-oxidants such as propylgallate seen intact cells. Glyceryl trihexadecanoate-l-C has been prepared in a maimer analogous to the syndiesis of dipalmitin (Dauben, 1948). Intermediates in the synthesis are palmitic-l-G aci methylhexadecanoate-l-G, and pal toyl-l-G chloride. Good yields of the glyceryl esters of hexadecanoic-l-G acid have also been obtained by Weinman et oL (1950) who used a modification of the method of Feuge et al. (1945). Weinman et dL (1951) prepared the glyceryl esters of octadecanoic-1-C and -6-G adds in the same way.
