Castanospermine s antitumor potential continues to receive attention. Recent in vitro studies have examined inhibition of tyrosinase activity in human melanoma cells (181), effects on intracellular transport of an avian erythroblastosis oncoprotein (277), and consequences for biosynthesis, maturation, and transport of ai-antitrypsin in the human hepatoma HepG2 cell line (257,272). The latter study was the first to show the inability of 239 to permeate the plasma membrane in intact HepG2 cells. In vivo experiments with nude mice proved that the alkaloid altered the glycosylation of endothelial cells, prevented angiogenesis, and inhibited tumor growth (273). However, recent in vivo studies failed to reveal cytotoxicity towards two rat prostate adenocarcinoma cell lines, or effects on cell characteristics related to metastatic potential (274). Uptake and metabolism of the more lipophilic 6-0-butanoylcastanospermine in tumor cell lines and after oral administration to mice was traced with C-labeled material, and showed rapid conversion into the parent alkaloid, which is undoubtedly the active metabolite (275). Multiple dosing in mice produced additive results. [Pg.142]

Serum TAG was significantly reduced by tlO,cl2-CLA as noted earlier. This is in agreement with our previous finding that the tlO,cl2-CLA isomer reduced apolipoprotein B secretion in cultured hepatoma HepG2 cells (38). It is worth noting that dietary CLA resulted in the inhibition of liver microsomal TAG transfer protein (MTP) activity (15). Taken together, the combination of the reduced TAG mass... [Pg.359]

A single mRNA species of 3.7 kb has been detected in human placenta and endothelial cell by Northern blot analysis of poly(A) " RNA (35). Thrombomodulin mRNA was not detected in hepatoma HepG2 cells or the monocytic U937 cells (35). [Pg.303]

Relatively little information is available. Savas et al. [1539] reported that P450 4A22 was only expressed at low levels in human hepatoma HepG2 cells and was refractory to treatment with the PPARa inducer Wyeth 14,643 or dexametha-sone, in contrast to P450 4A11. [Pg.613]

The only major study on regulation is work by Johnson and his associates [180] in hnman hepatoma HepG2 cells. The gene is regulated by the PPARa receptor, which regulates some other subfamily 4A P450s. [Pg.619]

MTBITC is the ITC derived from aliphatic GSL called glucoerucin and it is present in a large number of brassica vegetables in considerable amounts. It is of particular interest because it has been shown that SFN, which has been extensively studied and is one of the most potent chemopreventive ITC known to date, is oxidized in vivo to its structure analogue to MTBITC (Velasco et al. 2011a). Lamy and Mersch-Sun-dermann (2009) showed for the first time the inhibitory potency of this compound on the proliferation of human hepatoma HepG2 cells. [Pg.654]
