Thyroid epithelial cells synthesize and secrete T4 and T3 and make up the functional units of thyroid glandular tissue, the thyroid follicles. Thyroid follicles are hollow vesicles formed by a single layer of epithelial cells that are filled with colloid. T4,T3, and iodine are stored in the follicular colloid. T4 and T3 are derived from tyrosyl residues of the protein thyroglobulin (Tg). Thyroid follicular cells synthesize and secrete Tg into the follicular lumen. Thyroid follicular cells also remove iodide (I ) from the blood and concentrate it within the follicular lumen. Within the follicles, some of the tyrosyl residues of Tg are iodinated, and a few specific pairs of iodoty-rosyl residues may be coupled to form T4 and T3. Thus, T4, T3, and iodine (in the form of iodinated tyrosyl residues) are found within the peptide structure of the Tg that is stored in the follicular lumen. [Pg.743]

Blood is delivered to the human kidney by the renal artery. Blood flows to the kidneys of the adult human at a rate of roughly 1 L/min. The adult human kidney contains approximately 1 million functional units, called nephrons, to which the blood is delivered for removal of solutes. The functional unit is the reference point to which environmental impacts are attributed. The choice of functional unit depends on the application of the LCA. Erythromycins bind reversibly with a single high-affinity site on the 50S subunit of susceptible bacterial ribosomes. The site appears to be proteins L-15 and L-16, two of the 34 proteins constituting the ribosomal protein mass of the 50S unit. Removal of several L-16 proteins (by LiCl extraction) from a 50S subunit eliminates its affinity for EM peptidyl transfer ability is also eliminated. Restoring the L-16 protein alone reestablishes both functions. By itself L-16 has no EM binding capacity L-15 possesses both the capacity to bind EM and to effect peptidyl transfer, participated in some way by L-16. Both events occur on the P-site. Whether the bacteriostatic antimicrobial action of EM is due to the drugs inhibition of peptide bond formation or by the prevention of its translocation following peptide formation has not been established. To clarify the picture somewhat, perhaps it should be pointed out which aspects of protein synthesis are not affected by EM. They are amino acid activation, synthesis of the amino acid /RNA derivative, ribosomal association with raRNA, and reassociation of the 30S and 50S subunits to the complete ribosome. [Pg.258]
