Enzymatic relation

Figure 24.2a shows dual fluorescence intensity trajectories simultaneously recorded from a donor-acceptor labeled T4 lysozyme in the presence of substrate at pH 7.2. The anticorrelated fluctuations (Fig. 24.2a and b) are due to spFRET, reporting the donor-acceptor distance change associated with the protein conformational motion. Likewise, fluorescence trajectories of donor-acceptor labeled T4 lysozyme without substrates did not show anticorrelated behavior (Fig. 24.2c and d). We attribute this conformational motion to an enzymatic-related motion, most likely the open-closed hinge-bending motion... [Pg.474]

Many complex systems have been spread on liquid interfaces for a variety of reasons. We begin this chapter with a discussion of the behavior of synthetic polymers at the liquid-air interface. Most of these systems are linear macromolecules however, rigid-rod polymers and more complex structures are of interest for potential optoelectronic applications. Biological macromolecules are spread at the liquid-vapor interface to fabricate sensors and other biomedical devices. In addition, the study of proteins at the air-water interface yields important information on enzymatic recognition, and membrane protein behavior. We touch on other biological systems, namely, phospholipids and cholesterol monolayers. These systems are so widely and routinely studied these days that they were also mentioned in some detail in Chapter IV. The closely related matter of bilayers and vesicles is also briefly addressed. [Pg.537]

Protonated and diprotonated carbonic acid and carbon dioxide may also have implications in biological carboxylation processes. Although behavior in highly acidic solvent systems cannot be extrapolated to in vivo conditions, related multidentate interactions at enzymatic sites are possible. [Pg.197]

Biochemistry resulted from the early elucidation of the pathway of enzymatic conversion of glucose to ethanol by yeasts and its relation to carbohydrate metaboHsm in animals. The word enzyme means "in yeast," and the earfler word ferment has an obvious connection. Partly because of the importance of wine and related products and partly because yeasts are relatively easily studied, yeasts and fermentation were important in early scientific development and stiU figure widely in studies of biochemical mechanisms, genetic control, cell characteristics, etc. Fermentation yeast was the first eukaryote to have its genome elucidated. [Pg.366]

The enzymatic hydrolysates of milk casein and soy protein sometimes have a strong bitter taste. The bitter taste is frequently developed by pepsin [9001 -75-6] chymotrypsin [9004-07-3] and some neutral proteases and accounted for by the existence of peptides that have a hydrophobic amino acid in the carboxyhc terminal (226). The relation between bitter taste and amino acid constitution has been discussed (227). [Pg.296]

AH cephalosporins found in nature (Tables 1 and 2) have the D-a-aminoadipic acid 7-acyl side chain (21). AH of these compounds can be classified as having rather low specific activity. A substantial amount of the early work in the cephalosporin area was unsuccessfiiHy directed toward replacing the aminoadipic acid side chain or modifying it appropriately by fermentation or enzymatic processes (6,22). A milestone ia the development of cephalosporins occurred in 1960 with the discovery of a practical chemical process to remove the side chain to afford 7-ACA (1) (1). Several related processes were subsequendy developed (22,23). The ready avaHabHity of 7-ACA opened the way to thousands of new semisynthetic cephalosporins. The cephalosporin stmcture offers more opportunities for chemical modification than does that of penicillins There are two side chains that especiaHy lend themselves to chemical manipulation the 7-acylamino and 3-acetoxymethyl substituents. [Pg.21]

Copper is required for all forms of aerobic and most forms of anaerobic life. In humans, the biological function of copper is related to the enzymatic action of specific essential copper proteins (66). Lack of these copper enzymes is considered a primary factor in cerebral degeneration, depigmentation, and arterial changes. Because of the abundance of copper in most human diets, chemically significant copper deficiency is extremely rare (67). [Pg.212]

Citrate synthase catalyzes the metabolically important formation of citrate from ace-tyl-CoA and oxaloacetate [68]. Asp-375 (numbering for pig CS) has been shown to be the base for the rate-limiting deprotonation of acetyl-CoA (Fig. 5) [69]. An intennediate (which subsequently attacks the second substrate, oxaloacetate) is believed to be formed in this step the intermediate is thought to be stabilized by a hydrogen bond with His-274. It is uncertain from the experimental data whether this intermediate is the enolate or enol of acetyl-CoA related questions arise in several similar enzymatic reactions such as that catalyzed by triosephosphate isomerase. From the relative pK values of Asp-375... [Pg.232]

Preparation of PhAcOZ amino acids proceeds from the chloroformate, and cleavage is accomplished enzymatically with penicillin G acylase (pH 7 phosphate buffer, 25°, NaHS03, 40-88% yield). In a related approach, the 4-ace-toxy derivative is used, but in this case deprotection is achieved using the lipase, acetyl esterase, from oranges (pH 7, NaCl buffer, 45°, 57-70% yield). [Pg.543]

Hie ester linkage of aliphatic and aliphatic-aromatic copolyesters can easily be cleaved by hydrolysis under alkaline, acid, or enzymatic catalysis. This feature makes polyesters very attractive for two related, but quite different, applications (i) bioresorbable, bioabsorbable, or bioerodible polymers and (ii) environmentally degradable and recyclable polymers. [Pg.27]

For further discussion and for calibration purposes we will have to define a reference solution reaction which would be sufficiently related to the given enzymatic reaction. This will help us in removing the problems associated with concentration effect and in focusing on the actual catalytic advantage of the enzyme. To do this we describe the reference reaction in solution as... [Pg.139]

Enzymatic activities. The hydrolysis of ATP by actin-activated myosin is the characteristic enzymatic activity of muscle, smooth muscle included. All forms of smooth muscle myosin are slower than those of other muscles. The binding site for ATP and a reduced enzymatic activity are still present in monomeric myosin. The enzymatic activity of monomeric myosin is altered by a conformational change, (the 10S-6S transition) and the species of cations present in the reaction mixture. These differences relate to the possible mechanisms of regulation. [Pg.171]

The microsomal ethanol oxidizing system is another mechanism of ethanol metabolism. CYP2E1 may be an important enzyme in the metabolism of ethanol in heavy drinkers, who may have a 10-fold increase in activity. Two aUehc variants in the gene cl and c2) are associated with differing enzymatic activity. Approximately 40% of Japanese have the more active c2 allele, which is rare in individuals of European heritage (Sun et al. 2002). It is not believed to be a risk or protective factor in the development of alcohohsm, although current studies are examining its relationship to a variety of ethanol-related diseases. [Pg.8]

A Box-Behnken design was employed to investigate statistically the main and interactive effects of four process variables (reaction time, enzyme to substrate ratio, surfactant addition, and substrate pretreatment) on enzymatic conversion of waste office paper to sugars. A response surface model relating sugar yield to the four variables was developed on the basis of the experimental results. The model could be successfully used to identify the most efficient combination of the four variables for maximizing the extent of sugar production. [Pg.121]
