Drought also has a profound effect on protein synthesis. In many plant tissues, a reduced water potential causes a reduction of total protein synthesis and a rapid dissociation of polyribosomes. The latter has been shown not to be the consequence of increase in ribonuclease activity (Hsiao, 1973 Dhindsa Bewley, 1976). For a specific protein, Jacobsen, Hanson Chandler (1986) have shown in barley leaves that water stress enhances the synthesis of one of the a-amylase isozymes. The free fatty acid uptake by tissues is related directly to the plasma free fatty acid concentration, which in turn is determined by the rate of lipolysis in adipose tissue. After dissociation of the fatty acid-albumin complex at the plasma membrane, fatty acids bind to a membrane tty acid transport protein that acts as a transmembrane cotransporter with Na. On entering the cytosol, free fatty acids are bound by intracellular fatty acid-binding proteins. Direct evidence for oenocytes biosynthesizing hydrocarbon has come recently with the dissociation of oenocytes from epidermal cells and in vitro incubation with labeled propionate [233]. Differential uptake of some hydrocarbons in different tissues has also been documented although the exact mechanism behind the differential placement of hydrocarbons is unknown [20,128,230,232,234]. In the past, dissociation of the nucleoprotein complex has been brought about by salt solutions or by heat denaturation,129 but, more recently, decomposition has been effected by hydrolysis with trypsin,126 or by the use of dodecyl sodium sulfate130 or strontium nitrate.131 Some virus nucleoproteins are decomposed by ethyl alcohol.132 This effect may be similar to that of alcohol on the ribonucleoproteins of mammalian tissues. If minced liver is denatured with alcohol, and the dried tissue powder is extracted with 10% sodium chloride, the ribonucleoproteins are decomposed to give a soluble sodium ribonucleate while the deoxyribonucleoproteins are unaffected.133 On the other hand, extraction with 10 % sodium chloride is not satisfactory unless the proteins have first been denatured with alcohol. Denaturation also serves to inactivate enzymes of the tissues which might otherwise bring about degradation of the nucleic acid during extraction. [Pg.309]

Manganese(II)-A/, A/r -dipyridoxylethylenediamine-A/r, AT-diacetate 5,5 -bis(phosphate) 75 (DPDP) is clinically used for enhancing contrast in the liver (detection of hepatocellular carcinomas) (312). Some dissociation of Mn(II) appears to occur in the liver, and enhancement can also be obtained in functional adrenal tissues (313). Manganese(II)-tetrasulfonated phthalocyanine also shows tumor localization properties and is a more efficient relaxation agent than the analogous Gd(III) complexes (314). [Pg.238]

Prior to immunohistochemical staining, paraffin sections must be properly mounted onto slides, and then deparaffinized and rehydrated. To help adherence to the glass and decrease the chances of sections dissociating from the slides, paraffin tissue sections should be mounted on tissue-adhesive-coated slides. Last, chemical dissociation is commonly used in conjunction with mechanical or enzymatic procedures.
