The Perkin-Ehner Autosystem GC consists of a dual-injector port, dual-capillary-column configuration, and dual detectors [flame ionization (FID), electron-captnre (BCD)] connected via an analog-to-digital (A/D) interface to a central personal computer workstation which is driven by the Tnrbochrom (PE-Nelson) chromatography data processing software. Yon will enconnter two types of A/D interfaces in the laboratory. The 600 LINK interface provides for both data acquisition and instrnment control. The 900 interface provides for data acquisition only.

Measurement by gel permeation chromatography in 0.2 M phosphate buffer pH 7.0 with polystyrenesulfate or polyethylene glycols ([5] in the case of aureobasidium sp. A-91) as molecular weight standards. Data processing as de.scribed in Ref. [II]. 

Ethanol concentration in the fermentation broth is determined by using gas chromatography (HP 5890 series II with HP Chemstation data processing software, Hewlett-Packard, Avondale, PA) with a Poropak Q Column, and a Hewlett-Packard model 3380A integrator. A flame ionisation detector (FID) is used to determine ethanol. The oven temperature is maintained at 180 °C, and the injector and detector temperature are maintained at 240 °C. The sample taken from the fermentation media has to be filtered and any internal standard must be added for analysis based on internal standard methods otherwise, the area under the peak must be compared with known standard samples for calculation based on external standard methods. 

A representative example of this process is shown in Fig. 2. The spectra of the analyte peaks can be measured at the upslope, the top, and at the downslope, or the whole spectrum of the chromatography peak can be compared. In the latter case, the term totalpeak purity is used, and a purity curve of the peak can also be recorded. These operations can be performed by a HPLC system equipped with a DAD detector [13], or for TLC a densitometer that can measure the UV-Vis spectrum of the analyte spot should be used. If the value of the purity is 0.000 0.8900, it is not pure, and a purity of 0.9000-0.9500 means that the peak is contaminated (Shimadzu Class-VP, Chromatography Data System).

The Chem Master Workstation is a gas chromatography and gas chromatography-mass spectrometry data-processing system that speeds the flow of data through the laboratory and provides essential quality-assurance and quality-control review. It is a PC-based integrated hardware/ software system that converts gas chromatographic and gas chromatography-mass spectrometric data into reliable analytical reports. 

Note The abscissa of the TIC or RIC can either be plotted on the time scale, i.e., in units of seconds or minutes, or be labeled with scan numbers. Scan numbers are useful during data processing, while the time scale is better suited for comparison with other chromatography-mass spectrometry data. 

Many appHcations of chromatography have been automated to some degree attention has focused primarily on the automatic injection of samples or on data processing and reporting. However, the separation power of the column can be usefully exploited as a pretreatment process. 

Experimental Techniques. Chromatography was performed on a Varian model 5060 HPLC equipped with a RI-3 refractive index detector. A Vista Plus Gel Permeation Chromatography (GPC) data system was used consisting of a Vista 401 chromatography data system serially connected to an Apple II microcomputer. The Vista 401 performs data acquisition and allows data storage and automations capability while all SEC data processing is performed on the Apple II by means of user-interactive GPC software for automated, on-line calibration and polymer analysis.

Combination of the results with solvent extraction/liquid chromatography data may elucidate the role of free, unreacted monomer in the post-curing process. The main conclusions have already appeared elsewhere (9). Here we report in much more detail on the experimental techniques as well as on new results on the delay of shrinkage with respect to chemical conversion. 

In the paragraphs below, some of the above mentioned multivariate methods will be discussed in somewhat more detail, with respect to the data processing of signals obtained for hyphenated methods of the type chromatography-spectrometry and spectrometry-spectrometry.
