Broth, thioglycolate

For bacteriological analyses, the commercially available culture media were used Endo agar, elective saline agar, blood agar, thioglycolic medium and beef extract broth (BEB) with 1% glucose. Chocolate agar was used for the isolation of haemophilic flora. 

Media-hlled vials (soybean casein digest broth or fluid thioglycollate medium) are hlled on the hlling line, transported to the freeze drier, loaded into the chamber, subjected to a simulated lyophilization process, stoppered, sealed, and incubated for 14 days at 25°C. The minimum number of vials to be used is 5000. 

Soybean casein digest broth shall be used for normal media runs but thioglycolate broth shall be used for the detection of anaerobic organisms, especially when a filtered nitrogen purge is used to ensure anaerobic conditions. 

Before incubation of the vials, powder should be reconstituted with adapted media (TSB or thioglycolate broth) using aseptic technique under laminar flow. The reconstitution volume is according to the volume described in the original formula. Strict environmental monitoring should be followed through this step.

Remove duplicate 1 ml aliquots into a) thioglycollate broth, and b) Saboraud s liquid medium (Appendix 4). 

The next step is to gently rub the ulcer with a sterile calcium alginate swab moistened with thioglycolate broth or trypticase soy broth and inoculate three rows of C on each of the blood, chocolate, and Sabouraud s plates. Luminometer 3550i and Filtravette (New Horizons Diagnostics Corp., USA) were used for bioluminescent measurement. ATP-reagent (based on soluble Luciola mingrelica firefly luciferase) was developed in our laboratory. Dimethyl sulfoxide (DMSO) was from Reakhim (Russia). Nutrition broth (NB) from ICN, Tryptic Soy Broth (TSB) from Difco and Thioglycolate broth (TB) from Merck were used. Ultrapure deionized water was obtained on Milli-Q (Millipore). Broth culture of Escherichia coli LE392 was used in model experiments. 

Thioglycolate broth 3% thioglycolate broth (Becton Dickinson, Franklin Lakes, NJ) in sterile glass-distilled water. The bottle is placed in a boiling water bath to ensure complete dissolution of the powder. Autoclave the solution at 15 psi, 121°C for 15 min. Store at room temperature and use within 1 d.

The technique of membrane filtration is used whenever the nature of the product permits. The membrane is transferred to the growth medium, or the medium is transferred onto the membrane. Alternatively, the prepared sample is inoculated directly into the appropriate media. This method is only used when the product (e.g. some vaccines) cannot be dissolved or diluted in a nontoxic diluent. The media used are fluid thioglycolate medium (FTM) for aerobic, micro-aerophilic and anaerobic bacteria, and Soybean casein digest broth (SCDB) for aerobic bacteria and fungi. FTM and SCDB are incubated at 30-35 °C and 20-25 °C respectively, both for a period of not less than 14 days. This relatively long incubation period seems to be justified, because an unacceptable proportion of contaminants would be missed by limiting incubation to 7 days [60]. 

The processed pouches were subjected to sterility test as per (Indian Standard Institute 2168, Specification for Pomfret canned in oil. New Delhi, India). Commercial sterility is ensured so that there is no growth of microorganisms after thermal processing. Four pouches were incubated at 55°C for 4 days and another four were incubated at 37°C for 14 days before conducting the tests. The incubated pouches were aseeptieally opened and 1-2 g of the samples were taken with a sterilized forceps and inoeulated into the sterilized fluid thioglycolate broth in test tubes. In order to ereate anaerobie condition, a little sterilized liquid paraffin wax was put on the top of the broth and kept for incubation. The absence of any turbidity was observed after incubation, which indicated that the pouches were sterile. 

Assay Medium. Herrell el al. (16) recommend the use of fluid thioglycol-ate broth with the following composition yeast extract 5 g., Bacto-casitone 15 g., glucose 5 g., sodium chloride 2.5 g., Z-cystine 0.75 g., thioglycollic acid 0.3 ml., agar 0.75 g., resazurin (certified) 0.001 g., and distilled water 1 liter the final pH of this medium should be 7.1. 

The usual aerobic medium is plain nutrient broth, adjusted so that it has a final pH value after sterilisation of 7 2-7 8. The anaerobic medium is the same nutrient broth with the addition of 0 5 per cent cystine, 0 05-0 07 per cent agar, 0 05 per cent glucose, 0 05 per cent sodium thioglycollate and 0 1 per cent of a 1 in 1,000 aqueous solution of resazurin, and adjusted so that the final pH value after sterilisation is between 7-2 and 7 8. Both media may be filled in about 50-ml or 250-ml amounts into tubes plugged with cotton wool or into screw-capped bottles if bottles are used, they should be of such a size that they are not more than two-thirds filled with the aerobic medium, but for the anaerobic medium they should be filled to the neck to minimise the access of oxygen. If the anaerobic medium shows a pink colour extending to more than about one inch below the surface it should be heated in flowing steam for twenty to thirty minutes, and subsequently cooled immediately prior to use.
