Another peculiarity of the study is that the use of a biological system has allowed the authors to hypothesize a possible mechanism of action of the leachate as a mixture, hypothesis that could have been drafted on the basis of the only knowledge derived by chemical analysis. Researchers suggest that leachate inhibits cell proliferation at low doses probably inducing a reversible cell cycle arrest that becomes irreversible at high doses, probably due to leachate-induced oxidative stress. This activity is mainly due to the chemical compounds extracted in the aqueous phase. Similar effects were noticed by previous investigations on other human cells (human peripheral blood lymphocytes and a human breast cancer cell line, MCF-7) [31, 32], supporting the hypothesis that cells that survive the initial insult from leachate constituents maintains the potential to proliferate until the effects on cell metabolism lead to death. [Pg.180]

In contrast to P-gp and the MRP proteins, the breast cancer resistance protein (BCRP) contains six transmembrane domains and only one ATP-binding domain. It was first cloned from the breast cancer cell line MCF-7 selected in doxombicin, in the presence of the P-gp inhibitor verapamil. It is found in many human tissues, such as the placenta, small intestine, colon, and liver [133], It is localized to the apical membrane of epithelial cells of the small intestine and colon and to the bile canalicular membrane in the liver and is involved in reducing intestinal uptake, increasing hepatobiliary excretion, etc., leading to diminished oral bioavailability. cDNA sequences identical to BCRP and named MXR and ABCP, respectively, were independently isolated from human colon carcinoma cells and human placenta [134], BCRP requires... [Pg.383]

Toward the above objectives, and also that of developing new methods for proteomic investigation, this laboratory has undertaken the identification of proteins whose abundances or primary structures are modified in drug-resistant strains of the human breast cancer cell line MCF-7. The parental cell line and several strains selected by exposure to different anti-cancer drugs were provided by K. H. Cowan (Eppley Cancer Center, University of Nebraska), P. Gutierrez and D. D. Ross, (Greenebaum Cancer Center, University of Maryland), and J. A. Moscow, (Department of Pediatrics, University of Kentucky). [Pg.243]

The effect of various peptides and growth factors in small cell lung cancer cells has been reported by Bepler et al [121]. Four peptides isolated from completely different sources with structural similarities represent members of a new family of growth factors [121]. These peptides include 1) a 60 amino acid residue breast cancer- associated pS2 peptide isolated from human gastric juice and the culture media of the human breast cancer cell line MCF-7, 2) a 106 amino acid residue pancreatic spasmolytic polypeptide (PSP) isolated from porcine pancreas and 3) a 49 and 50 amino acid residue peptide predicted from a cyclic DNA isolated from the skin of frog, Xenopus laevis. [Pg.805]

Gokhale et al. [72] have synthesized a series of hydroxynaphthoquinone metal complexes as antitumor agents. The cytotoxic studies against the human breast cancer cell line MCF-7 revealed enhanced activities for the metal complexes. The highest activity is observed for the copper compound of lawsone, Fig. (12) (2-hydroxy-l,4-naphthoquinone). [Pg.314]

Lunatusin, an antifungal peptide (Mr 7 kDa) isolated from the seeds of Chinese lima bean [Phaseolus lunatus L.). The trypsin-stable peptide exerted an antifungal activity towards various fungal species, and also inhibited proliferation in the breast cancer cell line MCF-7 [J. H. Wong, T. B. Ng, Peptides 2005, 26, 2086]. [Pg.208]
