Polyphenol oxidase occurs within certain mammalian tissues as well as both lower (46,47) and higher (48-55) plants. In mammalian systems, the enzyme as tyrosinase (56) plays a significant role in melanin synthesis. The PPO complex of higher plants consists of a cresolase, a cate-cholase and a laccase. These copper metalloproteins catalyze the one and two electron oxidations of phenols to quinones at the expense of 02. Polyphenol oxidase also occurs in certain fungi where it is involved in the metabolism of certain tree-synthesized phenolic compounds that have been implicated in disease resistance, wound healing, and anti-nutrative modification of plant proteins to discourage herbivory (53,55). This protocol presents the Triton X-114-mediated solubilization of Vida faba chloroplast polyphenol oxidase as performed by Hutcheson and Buchanan (57). [Pg.186]

Outlined in this section will be the method of Maruyama et al. (12), who were successful in observing metaphase chromosomes inmeristem-atic cells of Vida faba when the fixation of freeze-fractured roots was performed after incubation in buffer solutions. The method allows for experimental studies on chromosome morphology. In fact, the published report also gives the effects of divalent cations on chromosome morphology. [Pg.294]

Fava beans (broad beans, Vida faba) contain a significant amount of L-DOPA. Anecdotal reports have noted improvement of Parkinson s disease after the consumption of fava beans (Spengos and Vassilopoulos 1988). Rabey and colleagues (1992,1993) have demonstrated the efficacy of fava beans in this respect. [Pg.205]

Anthraquinone glycosides found in senna (Cassia fistulosa) and Aloe spp. have been included in some commercial cathartics. Vicine is a glycoside in fava beans (Vida faba), and causes hemolytic anemia in people who have a genetic deficiency of glucose-6-phosphate dehydrogenase activity in their red blood cells. Fava beans are grown as a protein supplement for livestock. [Pg.54]

Aminouracil produces a block in the mitotic cycle of various plants. For example, cessation of mitosis occurred in Vida faba roots incubated 24 h with this compound [392—394]. Depending on different experimental conditions, thymidine or thymidylic acid may or may not alleviate these effects [392—394]. It was concluded that 5-aminouracil depressed the rate of DNA synthesis, which led to an accumulation of cells in the S phase. After removal of the agent, DNA synthesis resumed. Similar results have been observed with Allium cepa and Haplopappus gracilis [395, 396]. Inhibition of guanosine incorporation into RNA of meristematic cells in Vida faba by 5-aminouracil was also reported [397]. [Pg.307]

To investigate mass transfer of a radioactive vapour for which all surfaces are perfect sinks, Chamberlain (1966, 1974) used a vapour of 212Pb (ThB). This was generated by decay of 220 Rn (thoron) in a dust-free atmosphere and carried with a flow of filtered air into a wind tunnel (Fig. 6.2). Experiments with real and artificial grass swards in the tunnel are described below. In experiments with bean plants (Vida faba) in the tunnel, the deposition of 212Pb to individual leaves was measured, and analysed in terms of the Sherwood number, using the maximum chord of the leaves as the characteristic length, L. [Pg.197]

Earlier, we analyzed oat roots after BOA incubation of seedlings for at least 24 h and up to 72 h. Two new products were present in the extracts, which were identified as BOA-6-OH and BOA-6-O-glucoside.83 In addition, a third product was found that increased with incubation time. It was slightly more hydrophobic than BOA-6-OH with an UV scan very similar to BOA. The compound was purified, subjected to chemical identification and characterized as BOA-A/-glucoside. This product was not found in roots of Vida faba var. Alfred, but this seems to depend on variety e.g., Vida faba var. Dreifach Weisse, was able to produce the compound. Both varieties contained BOA-6-OH and the corresponding glucoside. [Pg.99]

ATPase activity was also studied by Friebe et al. in 1997.17 They correlated the BOA and DIBOA effects on radicle elongation of Avena sativa seedlings with their effects on the activity of plasma membrane H+-ATPase from roots of Avena sativa cv. Jumbo and from Vida faba cv. Alfred. They hypothesized that an alteration in the plasma membrane ATPase activity could be the reason for an abnormal nutrient absorption in plants exposed to hydroxamic acids, because of the role that this enzyme plays in the ion gradient and, therefore, in the ionic transport through plasma membrane. The results of this experiment showed a strong inhibition in the activity of this enzyme in the plasma membrane of chloroplast and mitochondria when it was exposed to BOA and DIMBOA. This alteration implies early interactions with the assayed hydroxamic acids. [Pg.255]
