Pharmacokinetics and toxicity have been identified as important causes of costly late-stage failures in drug development. Hence, physicochemical as well as ADMET properties need to be fine-tuned even in the lead optimization phase. Recently developed in silica approaches will further increase model predictivity in this area to improve compound design and to focus on the most promising compounds only. Abstract Phthalates are chemicals that have been used for over 80 years in large quantities due to their wide range of applications, mainly in the plastic industry. For many years, these compounds were not considered dangerous for humans due to their low toxicity shown in the preliminary studies and their low persistence. However, research conducted in recent years has evidenced their activity as endocrine disrupters, and they are now considered as emerging contaminants and included in the priority list of dangerous substances in the legislation of many countries. This chapter provides an overview on the properties, major uses, emission sources, environmental and human levels, current legislation, behavior and fate of phthalates, and their metabolites, with special emphasis on their toxicity and human exposure. [Pg.307]

Abiotic transformation of contaminants in subsurface natural waters result mainly from hydrolysis or redox reactions and, to lesser extent, from photolysis reactions. Complexation with natnral or anthropogenic ligands, as well as differential volatilization of organic compounds from multicomponent hquids or mixing with toxic electrolyte aqueous solutions, may also lead to changes in contaminant properties and their environmental effects. Due to its unique characteristics and physicochemical properties such as being less toxic, nonflammable, and having the extraction power tuned by temperamre and pressure, SC CO2 can be used as a green solvent for extraction of substances especially from solid or liquid substrates. Such extraction has been carried out on commercial scale for more than two decades and applications like decaffeination of coffee beans and black tea leaves and hops extraction are involved in large-scale processes [17]. Other extractions such as extraction of flavors, spices, and essential oils from plant materials are under investigation. The root of the plant Cryptolepis sanguinolenta (Lindl.) Schlt (Periplocaceae) is used in traditional African medicine to treat a variety of diseases including malaria, jaundice, hepatitis, urinary tract infections, hypertension, inflammatory conditions and stomach ache. Extracts of the roots are also used as a tonic often taken daily for years without evidence of toxicity. Various studies indicate that the crude extracts as well as the isolated alkaloidal constituents of the plant possess a number of interesting pharmacological properties. The focus of this overview is to highlight the potential of Cryptolepis sanguinolenta in modem health care. [Pg.231]
