Tablet wetting agents

Surfactants are used in capsule and tablet formulations as wetting agents to aid dissolution. 

Lubricants based on fatty acids, because of their low water solubility, are unsuitable for tablets which must be dissolved in water before use. Polyethylene glycol 6000 (macrogol 6000) is soluble in water, but its lubricant activity is limited. Magnesium lauryl sulfate has been suggested as a water-soluble substitute for magnesium stearate. In addition to its lubricant action, this substance, like sodium lauryl sulfate, is an effective wetting agent. ... 

Addition of wetting agents such as sodium lauryl sulfate or sodium docusate can assist this water penetration by lowering the surface tension, and they are often used in conjunction with hydrophobic lubricants such as magnesium stearate (see the article on Tablet Disintegrants and Disintegration in this encyclopedia). 

Hypromellose phthalate can be applied to tablet surfaces using a dispersion of the micronized hypromellose phthalate powder in an aqueous dispersion of a suitable plasticizer such as triacetin, triethyl citrate, or diethyl tartrate along with a wetting agent. ... 

In tableting, polydextrose solutions are used as binders in wet-granulation processes. Polydextrose is also used in the manufacture of directly compressible tableting excipients. Polydextrose solutions may also be used, in conjunction with other materials, as a film and tablet coating agent.

Zein is used as a tablet binder in wet-granulation processes or as a tablet-coating agent mainly as a replacement for shellac. It is used primarily as an enteric-coating agent or in extended-release oral tablet formulations. Zein is also used in food applications as a coating agent. See Table I. 

Poloxamers are used as emulsifying agents for intravenous fat emulsions, as solubilising agents to maintain clarity in elixirs and syrups, and as wetting agents for antibacterials. They may also be used in ointment or suppository bases and as tablet binders or coaters. 

The performance requirements of a tablet were discussed in the previous section, and it is clear that most tablets will contain several ingredients. The drug substance, probably a filler, a disintegrant and a lubricant will be common to most formulations, while glidants, colourants and wetting agents may also be included. The quality of the final product will be as dependent on the way in which the components are combined as it is on the components selected. Traditionally, tablet formulations have been prepared by one of two methods direct compression or granulation.

Figure 9.14 shows how magnesium stearate retards the dissolution of salicylic acid from tablets whereas tablets prepared using sodium lauryl sulfate (a soluble hydrophilic wetting agent) as a lubricant had rapid dissolution.
